The first evident break in left-right symmetry of the primitive zebrafish heart tube is shift pattern BMP4 expression from radially symmetric to left-predominant. midline then 'jogs' left and subsequently loops right. We examined 279 mutations, affecting more than 200 genes, found 21 mutations that perturb this process. Some cause remain symmetric. Others randomize asymmetric pattern. Retention associated with failure jog: right-predominance reversal direction jogging looping. Raising...
To determine whether an angiotensin-converting enzyme (ACE) inhibitor, enalapril, prevents cardiac function deterioration (defined using maximal index [MCI] on exercise testing or increase in left ventricular end-systolic wall stress [LVESWS]) long-term survivors of pediatric cancer. This was a randomized, double-blind, controlled clinical trial comparing enalapril to placebo 135 cancer who had at least one abnormality identified any time after anthracycline exposure. There no difference the...
We have identified and characterized a zebrafish recessive maternal effect mutant, ichabod, that results in severe anterior dorsal defects during early development. The ichabod mutation is almost completely penetrant, but exhibits variable expressivity. All mutant embryos fail to form normal embryonic shield; most head notochord excessive development of ventral tail fin tissue blood. Abnormal patterning can first be observed at 3.5 hpf by the lack nuclear accumulation (beta)-catenin yolk...
ABSTRACT Transgenic mice were generated containing a cytomegaloviral promoter driven construct (CMV43) expressing the gap junction polylpeptide connexin 43. RNA and protein analysis confirmed that transgene was being expressed. In situ hybridization of embryo sections revealed expression targeted to dorsal neural tube in subpopulations crest cells. This pattern identical seen transgenic harboring other constructs by (Kothary, R., Barton, S. C., Franz, T., Norris, M. L., Hettle, Surani, A. H....
We have cloned and sequenced a zebrafish (Danio rerio) Vascular Endothelial Growth Factor (vegf) cDNA. It encodes precursor protein of 188 amino acids with putative 23 signal peptide. Sequence comparison analysis indicates that the vegf cDNA corresponds to human VEGF165 isoform shows about 52% identity at acid level. A 2.8 kb message RNA was detected in adult by Northern blot analysis. Expression vegf165 is also RT-PCR fish throughout embryonic development. Whole mount situ hybridization...
Interrupted aortic arch (IAA) is often related developmentally to subaortic obstruction (SAO). When severe, SAO must be addressed in surgical management of IAA. From 1990 1993, 25 neonates presented for initial IAA complexes. Associated lesions were ventricular septal defect (VSD) with or without atrial (19 patients), truncus arteriosus (3 tricuspid atresia transposition the great arteries (1 patient), VSD and d-transposition patient). Overall hospital mortality was 20% (five deaths). One...
