目的 探讨硫化镉量子点(CdS quantum dots,CdS QDs)对中国仓鼠肺成纤维细胞(CHL)的毒性作用及其可能的作用机制.方法 实验设2.5、5.0、10.0、20.0、50.0、100.0μg/ml CdS QDs(乳化膜法合成)6个剂量组作用于CHL细胞,对照组予等量的生理盐水.用荧光倒置显微镜观察细胞的形态学变化,噻唑蓝(MTT)法检测CdS QDs对CHL细胞增殖的抑制作用.CHL细胞分别暴露于不同浓度的CdS QDs 24 h后.测定细胞内乳酸脱氢酶(LDH)释放率及丙二醛(MDA)、谷胱甘肽(GSH)含量.结果 QDs染毒24 h后,细胞变形、收缩,胞体折光性脆弱.随着CdS QDs染毒剂量的增加,CHL细胞存活率逐渐下降,且各剂量组间与对照组相比,差异均有统计学意义(P<0.05或P<0.01).CdS h后,随染毒剂鼍的增加,各剂量组细胞LDH释放率均明显上升,差异有统计学意义(P<0.01).20.0、50.0μg/ml...
