RNA interference (RNAi) is an important antiviral defense response in plants and invertebrates; however, evidences for its contribution to mammalian are few. In the present study, we demonstrate anti-dengue virus role of RNAi cells. Dengue infection Huh 7 cells decreased mRNA levels host factors, namely, Dicer, Drosha, Ago1, Ago2, corollary, silencing these genes virus-infected enhanced dengue replication. addition, observed downregulation many known human microRNAs (miRNAs) viral infection....
Significance Atopic dermatitis (AD) is a chronic inflammatory skin condition affecting 13% of adults and 10% children worldwide. The inflammation seen in AD driven by type 2 cytokines, including IL-4 IL-13. We have an incomplete understanding about how these cytokines impact the epithelial barrier drive pathology. Our study demonstrates that IL-13 promote androgen production lipid abnormalities sebocytes. These findings deepen our immune system on epithelium.
Our previous studies consistently demonstrate enhanced pulmonary vascular remodeling in HIV-infected intravenous drug users, and simian immunodeficiency virus-infected macaques or HIV-transgenic rats exposed to opioids cocaine. Although we reported an associated increase perivascular inflammation, the exact role of inflammatory cells development remains unknown. In this study, cocaine (H+C)-treated human monocyte derived macrophages released a higher number extracellular vesicles (EVs),...
Intravenous drug use is one of the major risk factors for HIV-infection in HIV-related pulmonary arterial hypertension patients. We previously demonstrated exaggerated vascular remodeling with enhanced apoptosis followed by increased proliferation endothelial cells on simultaneous exposure to both opioids and HIV protein(s). Here we hypothesize that exacerbation autophagy may be involved switching from an early apoptotic state later hyper-proliferative state. Treatment human microvascular...
Background Earlier, we reported that the simultaneous exposure of pulmonary arterial smooth muscle cells to HIV proteins and cocaine results in attenuation antiproliferative bone morphogenetic protein receptor‐2 ( BMPR 2) expression without any decrease its mRNA levels. Therefore, this study, aimed investigate micro RNA ‐mediated posttranscriptional regulation 2 expression. Methods Results We identified a network targeting s including miR‐216a be upregulated response Tat‐mediated...
The host-mediated RNAi pathways restrict replication of viruses in plant, invertebrate and vertebrate systems. However, comparatively little is known about the interplay between various viral infections mammalian hosts. We show present study that siRNA-mediated silencing Drosha, Dicer Ago2 [argonaute RISC (RNA-induced complex) catalytic component 2] transcripts Huh7 cells resulted elevated levels HBV (hepatitis B virus)-specific RNAs and, conversely, we observed a decrease mRNA protein same...
We previously reported enhanced proliferation of smooth muscle cells on the combined exposure HIV proteins and cocaine leading to development HIV-pulmonary arterial hypertension. Here, we attempt comprehensively understand interactions between long noncoding RNAs (lncRNAs), mRNAs micro-RNAs (miRNAs) determine their role in hyperplasia. Differential expression lncRNAs, miRNAs were obtained by microarray small-RNA sequencing from HPASMCs treated with without and/or HIV-Tat. LncRNA mRNA...
Abstract Staphylococcus aureus, the most frequent cause of skin infections, is more common in men than women and selectively colonizes during inflammation. Yet, specific cues that drive infection these settings remain unclear. Here we show host androgens testosterone dihydrotestosterone promote S. aureus pathogenesis infection. Without secretion hormones, vivo limited. Testosterone activates virulence a concentration dependent manner through stimulation agr quorum sensing system, with...
microRNAs play important regulatory role in all intrinsic cellular functions. Amongst lepidopteran insects, miRNAs from only Bombyx mori have been studied extensively with a little focus on Spodoptera sp. In the present study, we identified total of 226 frugiperda cell line Sf21. Of total, 116 were well conserved within other like B. mori, Drosophila melanogaster and Tribolium castenum while remaining 110 as novel based comparative analysis insect miRNA data set. Landscape distribution Sf21...
Abstract Viruses encode multiple proteins that interact with different host factors to aid in their establishment inside the host. Viral Suppressor of RNA silencing (VSR) are one such class have been shown components machinery involved post transcriptional gene silencing, a known antiviral defence mechanism. In present study, we showed dengue NS3, VSR not only interacts HSPA1A, cellular chaperone, but also modulates its expression levels. Further, revealed HSPA1A associated through...
Introduction and hypothesis: Intravenous drug use (IVDU) is considered to be one of the important risk factors in development HIV-associated pulmonary arterial hypertension (HPAH). Our previous findings demonstrate that exposure cocaine HIV infected individuals has an additive effect enhancing proliferation smooth muscle cells, thus further amplifying arteriopathy. Furthermore, various studies suggest failure Hippo pathway increased YAP/TAZ activity (PH). Based on this, we hypothesized YAP...
