Antibody drug conjugates (ADCs) are an emerging class of treatments designed to improve efficacy and decrease toxicity compared with other systemic therapies through the selective delivery cytotoxic agents tumor cells. Datopotamab deruxtecan (Dato-DXd) is a novel ADC comprising topoisomerase I inhibitor payload monoclonal antibody directed trophoblast cell-surface antigen 2 (TROP2), protein that broadly expressed in several types solid tumors. Dato-DXd being investigated across multiple...
Abstract Objectives Cefepime-induced neurotoxicity (CIN) has been demonstrated to be associated with cefepime plasma concentrations; however, the toxicity threshold remains unclear. The primary objective of this study was identify trough concentration at which occurs. Secondary objectives were determine incidence CIN a large tertiary institution and patient factors development CIN. Methods A retrospective review all adult patients administered between October 2017 May 2018 in hospital...
Aims The purpose of the study was to assess status emerging therapeutic drug monitoring (TDM) anti‐infective agents in Australian hospitals. Methods A nationwide cross‐sectional survey all hospitals operating public and private health sector conducted between August September 2019. consisted questions regarding institutional TDM practice for clinical vignettes specific β‐lactam antibiotics. Results Responses were received from 82 unique institutions, representing states territories. All 29...
This study evaluated the ability of a pilot therapeutic drug monitoring (TDM) Advisory Service to facilitate vancomycin target attainment within real-world clinical setting. The provided area under concentration-time curve (AUC)-guided dose recommendations, using Bayesian forecasting software and expertise, prescribers at an Australian hospital. A retrospective audit intravenous therapy (> 48 hours) in adults (≥ 18 years old) was undertaken over 54-month period evaluate established...
Bayesian forecasting software can assist in guiding therapeutic drug monitoring (TDM)-based dose adjustments for amikacin to achieve targets. This study aimed evaluate prescribing and TDM practices, determine the suitability of model incorporated into DoseMeRx® as a replacement previously available (Abbottbase®).Patient demographics, pathology, dosing history, concentrations Abbottbase® predicted targets (area under curve up 24 hours, maximum concentration trough concentration) were...
Abstract: Ceftazidime–avibactam (CTZ-AVM) is a novel cephalosporin/beta-lactamase inhibitor with broad-spectrum activity against multidrug-resistant Pseudomonas aeruginosa . Ceftazidime-induced neurotoxicity well-described adverse effect, particularly in patients renal insufficiency. However, appropriate dosing of ceftazidime–avibactam undergoing replacement therapy (RRT) sparsely investigated, and therapeutic drug monitoring to guide remains lacking. Furthermore, when dose adjustment for...
We present the first published case of successfully treated disseminated Aspergillus lentulus infection in a solid organ transplant recipient with invasive pulmonary disease, endophthalmitis, and cerebral abscess. This highlights important challenges associated treating A. lentulus, particularly regarding antifungal resistance toxicities long-term therapy.
