Pyridin‐2‐yl‐ and 4,6‐dimethylpyrimidin‐2‐yl‐cyanamides entered into an alkylation reaction in the form of sodium salts. Pyridin‐2‐yl cyanamide 2 was alkylated at endo‐nitrogen atom pyridine ring, while 4,6‐dimethylpyrimidin‐2‐yl 1 effectively exo‐nitrogen amino group. The 3- R -thieno[3,2- d ]pyrimidine-2,4(1 H ,3 )-diones ( 5a- с ) and -2-thioxo-2,3-dihydrothieno[3,2- ]pyrimidin-4(1 )-ones 5d-g were synthesized using methyl 3-aminothiophene-2-carboxylate alkyl-, arylisocyanates isothiocyanates respectively, which in turn converted into its N- и S- alkyl derivates 8a-e, 9a-g ).  2-aminothieno[3,2- ]pyrimidin-4(3 )-one 14a-c , 16 received as a result the interaction  with benzoyl- pyrimidin-2-yl cyanamides.
