Abstract The clinical success of CRISPR therapies hinges on the safety and efficacy Cas proteins. Cas9 from Francisella novicida (FnCas9) is highly precise, with a negligible affinity for mismatched substrates, but its low cellular targeting efficiency limits therapeutic use.
