Summary High biomass crops have recently attracted significant attention as an alternative platform for the renewable production of high energy storage lipids such triacylglycerol ( TAG ). While typically accumulates in seeds compounds fuelling subsequent germination, levels vegetative tissues are generally low. Here, we report accumulation more than 15% (17.7% total lipids) by dry weight N icotiana tabacum (tobacco) leaves co‐expression three genes involved different aspects without...
Leucoanthocyanidin reductase (LAR) catalyzes the synthesis of catechin, an initiating monomer condensed tannin or proanthocyanidin (PA) synthesis, from 3,4-cis-leucocyanidin and thus is first committed step in PA biosynthesis. The enzyme was purified to near homogeneity PA-rich leaves legume Desmodium uncinatum (Jacq.) DC, partially sequenced corresponding cDNA cloned. identity confirmed by expressing active recombinant LAR Escherichia coli tobacco white clover. a 43 kDa (382 amino acids)...
Summary The anthocyanin and proanthocyanidin (PA) biosynthetic pathways share common intermediates until leucocyanidin, which may be used by leucoanthocyanidin dioxygenase (LDOX) to produce anthocyanin, or the enzyme reductase (LAR) catechin, a precursor of PA. Arabidopsis mutant tannin deficient seed 4 ( tds4‐1 ) has reduced PA level altered pattern accumulation. We identified TDS4 gene as LDOX complementation mutation either with cosmid encoding 35S:LDOX construct. Independent lines T‐DNA...
Summary Wheat streak mosaic virus (WSMV) is a persistent threat to wheat production, necessitating novel approaches for protection. We developed an artificial miRNA strategy against WSMV, incorporating five amiRNAs within one polycistronic amiRNA precursor. Using sequence and folding rules, we chose targeting conserved regions of WSMV but avoiding off‐targets in wheat. These replaced the natural each arms rice miR395, producing precursor, FanGuard (FGmiR395), which was transformed into...
Significance The aleurone, storing proteins, lipids, vitamins, and minerals, is the most nutritious part of cereal grains. Genetic analyses were conducted to screen for mutants with thickened identified thick aleurone 2-1 , which exhibits a multicell-layered an improved nutritional profile. Map-based cloning showed that TA2 encodes DNA demethylase. This study provides strategy enhancing value rice, possibly other cereals as well.
Abstract Proanthocyanidin (PA), or condensed tannin, is a polymeric flavanol that accumulates in number of tissues wide variety plants. In Arabidopsis, we found PA precursors (detected histochemically using OsO4) accumulate the endothelial cell layer seed coat from two-terminal stage embryo development onwards. To understand how made, screened mature pools T-DNA-tagged Arabidopsis lines to identify mutants defective synthesis and six tds(tannin-deficient seed) complementation groups...
Barley yellow dwarf virus (BYDV) resistance has been transferred to wheat from a group 7 chromosome of Thinopyrum (Agropyron) intermedium. The source the gene was L1 disomic addition line, which carries 7Ai-1 chromosome. locus is on long arm this BYDV resistant recombinant lines were identified after three or more generations selection against Th. intermedium short marker (red coleoptile) and for presence resistance. One line produced by ph. mutant induced homoeologous pairing 14 cell...
Summary Only plants of the Papaver genus (poppies) are able to synthesize morphinan alkaloids, and cultivation P. somniferum , opium poppy, remains critical for production supply morphine, codeine various semi‐synthetic analgesics. Opium poppy was transformed with constitutively expressed cDNA codeinone reductase ( PsCor1.1 ), penultimate step in morphine synthesis. Most transgenic lines showed significant increases capsule alkaloid content replicated glasshouse field trials over 4 years....
Potato tuber is a high yielding food crop known for its levels of starch accumulation but only negligible triacylglycerol (TAG). In this study, we evaluated the potential lipid production in potato tubers by simultaneously introducing three transgenes, including WRINKLED 1 (WRI1), DIACYLGLYCEROL ACYLTRANSFERASE (DGAT1) and OLEOSIN under transcriptional control tuber-specific (patatin) constitutive (CaMV-35S) promoters. This coordinated metabolic engineering approach resulted over 100-fold...
Zhong 5 is a partial amphiploid (2n = 56) between Triticum aestivum 42) and Thinopyrum intermedium carrying all the chromosomes of wheat seven pairs from Th. intermedium. Following further backcrossing to wheat, six independent stable 2n 44 lines were obtained representing 4 disomic chromosome addition lines. One confers barley yellow dwarf virus (BYDV) resistance, whereas two other carry leaf stem rust resistance; one latter also stripe resistance. Using RFLP isozyme markers we have shown...
Summary We demonstrate that both over‐expression and suppression of the gene encoding morphinan pathway enzyme salutaridinol 7‐ O ‐acetyltransferase (SalAT) in opium poppy affects alkaloid products accumulate. Over‐expression most transgenic events resulted an increase capsule morphine, codeine thebaine on a dry‐weight basis. The line with highest content had 41%, 37% 42% greater total alkaloids than control three independent trials over 3 years. DNA‐encoded hairpin RNA‐mediated SalAT novel...
Dual-purpose cereals have been important for increasing the flexibility and profitability of mixed farming enterprises in southern Australia, providing winter feed when pasture dry matter production is low, then recovering to produce grain. A perennial dual-purpose cereal could confer additional economic environmental benefits. We establish that, at end a second growth season, selected were able achieve up 10-fold greater below-ground biomass than resown annual wheat. review expand data on...
