Lipoprotein synthesis by the colonic adenocarcinoma cell line Caco-2 was investigated to assess utility of this as a model for in vitro study human intestinal lipid metabolism. Electron micrographic analysis conditioned medium revealed that under basal conditions culture post-confluent cells synthesize and secrete lipoprotein particles. Lipoproteins density (d) less than 1.063 g/ml consist heterogeneous population particles (diameter from 10 90 nm). This fraction consists very low...

Measurements of upper-air densities and temperatures from 12 flights the rocket-borne falling sphere are presented. They show little seasonal variation at 32° 38° N, but large variations in a few winter days 59°N. A low arctic density 60 km results linear latitude gradient about 2 per cent degree latitude. Such was measured survey. An abrupt warming 45 detected Fort Churchill January 1958 related to larger scope by balloons. The average air above March warmer 75 cooler 50 30 than 38°N.

Replication-deficient adenoviral vectors have been developed for the delivery of DNA sequences encoding a variety proteins intended management disease through gene therapy. One concern is occur- rence replication-competent adenovirus (RCA) in population replication-deficient as result recombination or contamination. To address this concern, it necessary to determine fre- quency occurrence and fully characterize molecular structure biological infectivity RCA. rAd/p53 pIX-deleted p53 therapy...

We have analyzed determinants of the synthesis and secretion apolipoproteins including mRNA for apolipoproteins, in human colon carcinoma cell line Caco-2 during differentiation continuous culture. Significant increases both cellular secreted apolipoprotein A-I were observed early process. Increases B limited to protein started later Levels A-I, A-IV, B, C-III, E increased significantly between time cells reached confluence 1 week postconfluence. The kinetics accumulation influenced by...

Apolipoprotein A-I (apoA-I) is the major protein constituent of plasma high density lipoproteins (HDL). To examine apoA-I processing and secretion, human gene (2.2-kilobase PstI-PstI fragment) linked to mouse metallothionein promoter was transfected by electroporation into NIH 3T3 fibroblasts along with plasmid pSV2 neo, which confers neomycin resistance. Transfected cells were selected for resistance screened ability produce enzyme-linked immunosorbent assay. In absence lipids in medium,...

Abstract In manufacturing monoclonal antibodies (mAbs), it is crucial to be able predict how process conditions and supplements affect productivity quality attributes, especially glycosylation. Supplemental inputs, such as amino acids trace metals in the media, are reported cell metabolism glycosylation; quantifying their effects essential for effective development. We aim present validate, through a commercially relevant culture process, technique modeling efficiently. While existing models...

Abstract The move to integrated continuous bioprocessing (ICB), while providing a means for process intensification, can put added strain on analytics when conventional methods are used. For instance, traditional microbial provide minimal value ICB processes given that the time required data become available is much longer than typical full cycle of manufacturing process. Although rapid detection has been in discussion over 30 years, it still not routinely deployed commercial...
