In the mouse fibroblast cell line C3H 10T1/2 and chicken DF1, ganglioside GM3 is major glycosphingolipid component of plasma membrane. Expression viral oncoprotein Jun (v-Jun) induces transformed clones with greatly reduced levels synthase (lactosylceramide α2,3-sialyltransferase) mRNA in both DF1 cultures. Compared nontransformed controls, v-Jun transfectants show enhanced ability anchorage-independent growth, their growth rates as adherent cells are increased. When gene transfected pcDNA...
Autoclaved mycelium of Aspergillus oryzae Y-2-32 adsorbed melanoidin, especially lower molecular weight fractions, and the degree adsorption was influenced by kind sugars utilized for growth. The melanoidin-adsorbing ability mycelia repressed a high concentration salt. Furthermore, it decreased to half initial level on washing with 0.1% Tween 80 solution entirely lost sodium dodecylsulphate solution.
Eukaryotic translation initiation factor 3 (eIF3) in the yeast Saccharomyces cerevisiae comprises about eight polypeptides and plays a central role binding of methionyl-tRNAi mRNA to 40S ribosomal subunit. The fourth largest subunit, eIF3-p39, was gel purified, 12-amino-acid tryptic peptide sequenced, enabling cloning TIF34 gene. encodes 38,753-Da protein that corresponds eIF3-p39 size antigenicity. Disruption is lethal, depletion by glucose repression expressed from GAL promoter results...
Translation initiation factor eIF1A is required in vitro for maximal rates of protein synthesis mammalian systems. It functions primarily by dissociating ribosomes and stabilizing 40 S preinitiation complexes. To better elucidate its precise role promoting the translation process, yeast form has been identified Saccharomyces cerevisiae purified to homogenity on basis cross-reaction with antibodies prepared against eIF1A. The apparent mass (22 kDa) resembles that homolog (20 kDa), active...
We evaluated electroporation as an alternative system for genetic exchange one of the coryneform bacteria, Brevibacterium flavum MJ233. The maximum number transformants, 6 x 10(4) cells, was obtained when cells were cultured with Penicillin G (1 U/ml) and harvested at middle-log phase. Electroporation done using 12.5 kV/cm pulse field strength, 1 10(10) microgram plasmid DNA. Other lactofermentum ATCC 13869, Corynebacterium glutamicum 31830, B. stationis IFO 12144 also transformed by...
Abstract Transfer of activated sugar‐nucleotides from the cytoplasm to lumen Golgi is an essential requirement for glycosylation glycoproteins, proteoglycans and glycosphingolipids. Although mannosylation major modification in yeast Saccharomyces cerevisiae , several reports suggest presence galactose residues on proteins sphingolipids. We have detected α‐galactosylated O ‐linked chitinase by lectin blotting cells that functionally express gma12 + gene, encoding α1,2‐galactosyltransferase...

Bioelectrochemical systems such as microbial fuel cells (MFCs) are promising new technologies for efficient removal of organic compounds from industrial wastewaters, including that generated swine farming. We inoculated two pairs laboratory-scale MFCs with sludge granules a beer wastewater-treating anaerobic digester (IGBS) or taken the bottom tank receiving wastewater (SS). The SS-inoculated MFC outperformed IGBS-inoculated regard to COD and VFA electricity production. Using metagenomic...
We have studied in vivo neo-galactosylation Saccharomyces cerevisiae and analyzed the critical factors involved this system. Two heterologous genes, gma12 + encoding α1,2-galactosyltransferase (α1,2 GalT) from Schizosaccharomyces pombe UGT2 UDP-galactose (UDP-Gal) transporter human, were functionally expressed to examine intracellular conditions required for galactosylation. Detection by fluorescence labeled α-galactose specific lectin revealed that 50% of cells incorporated galactose cell...
We developed a proprietary anion exchange membrane (AEM) for wastewater treatment as an alternative to commercial products. Following the successful development of hydrogel cation on porous ceramic support, we used same approach fabricate AEM. Different positively charged monomers and conditions were tested, all AEMs tested nitrate phosphate removal from buffers by electrodialysis. The best AEM was further with real swine electrodialysis in bioelectrochemical denitrification system (BEDS)....
