To investigate the nature of somatic von Hippel-Lindau (VHL) mutations, we analyzed 173 primary sporadic human renal cell carcinomas for mutations VHL tumor suppressor gene, using polymerase chain reaction (PCR) and single-strand conformational polymorphism analysis (SSCP) DNA. We detected abnormal SSCP pattern in 73 samples. After sequencing, identified microdeletions 58% cases, microinsertions 17%, nonsense 8%, missense 17%. Among these 50% correspond to new mutations. were found only...

Abstract The pathophysiological mechanism involved in side effects of radiation therapy and especially the role endothelium remains unclear. Previous results showed that plasminogen activator inhibitor-type 1 (PAI-1) contributes to radiation-induced intestinal injury suggested this could be driven by an endothelium-dependent mechanism. We investigated whether endothelial-specific PAI-1 deletion affect injury. created a mouse model with specific (PAI-1KO endo ) Cre-LoxP system. In...

Normal tissue toxicity still remains a dose-limiting factor in clinical radiation therapy. Recently, plasminogen activator inhibitor type 1 (SERPINE1/PAI-1) was reported as an essential mediator of late radiation-induced intestinal injury. However, it is not clear whether PAI-1 plays role acute damage and we hypothesized that may play the endothelium radiosensitivity. In vivo, model enteropathy −/− mice, apoptosis radiosensitive compartments, epithelial microvascular quantified. vitro,...

Abstract The efficacy and side effects of radiotherapy (RT) depend on parameters like dose the volume irradiated tissue. RT induces modulations tumor immune microenvironment (TIME) that are dependent dose. Low (LDRT, i.e., single doses 0.5–2 Gy) has been shown to promote infiltration into tumor. Here we hypothesize partial irradiation combining immunostimulatory/non-lethal properties LDRT with cell killing/shrinkage high (HDRT) within same mass could enhance anti-tumor responses when...

Radiation-induced lesion outcomes of normal tissues are difficult to predict. In particular, radiotherapy or local exposure a radioactive source by accident can trigger strong injury the skin. The finding biomarkers is fundamental relevance for prediction apparition and its evolution, settlement therapeutic strategies. order study radiation-induced cutaneous lesions, we developed mouse model in which dorsal skin was selectively exposed ionizing radiation (IR). 2-D difference gel...

Abstract Motivation: Identifying the set of genes differentially expressed along time is an important task in two-sample course experiments. Furthermore, estimating at which periods differential expression present can provide additional insight into temporal gene functions. The current detection methods are designed to detect difference observation intervals or on single measurement points, warranting dense measurements characterize full patterns. Results: We propose a novel Bayesian...

As it is altered by ionizing radiation, the vascular network considered as a prime target in limiting normal tissue damage and improving tumor control radiation therapy. Irradiation activates endothelial cells which then participate recruitment of circulating cells, especially overexpressing cell adhesion molecules, but also other yet unknown mechanisms. Since protein glycosylation an important determinant adhesion, we hypothesized that could alter pattern thereby impact cells. Herein, show...
