Abstract Multigene panel tests (MGPTs) revolutionized the diagnosis of Lynch syndrome (LS), however noncoding pathogenic variants (PVs) can only be detected by complementary methods including whole genome sequencing (WGS). Here we present a DNA-, RNA- and tumor tissue-based WGS prioritization workflow for patients with suspicion LS where MGPT no LS-related PV. Among 100 enrolled patients, 28 simple PVs an additional 3 complex PVs. 69 MGPT-negative lack somatic MLH1 promoter methylation in...

ABSTRACT Wine production is a complex procedure in which an important role played by many microorganisms, particularly yeasts and bacteria. In modern wineries, alcoholic fermentation usually carried out adding microbial starter cultures of Saccharomyces cerevisiae strains for precisely controlled production. Nowadays, the Slovak Republic, autochthonous vinification getting more popular. The present article deals with comparison two approaches, namely spontaneous standard commercial S....

To explore a genomic pool of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) during the pandemic, Ministry Health Slovak Republic formed genomics surveillance workgroup, and Public Authority launched systematic national epidemiological using whole-genome sequencing (WGS). Six out seven centers implementing Illumina technology were involved in SARS-CoV-2 virus program. Here we analyze total 33,024 isolates collected from population 1 March 2021, to 31 2022, that sequenced...

Summary Sensitive and accurate RT‐qPCR tests are the primary diagnostic tools to identify SARS‐CoV‐2‐infected patients. While many SARS‐CoV‐2 available, there significant differences in test sensitivity, workflow (e.g. hands‐on‐time), gene targets other functionalities that users must consider. Several publicly available protocols shared by reference labs public health authorities provide useful for diagnosis, but have shortcomings related sensitivity laborious workflows. Here, we describe a...

Abstract Inflammatory bowel disease (IBD) is a group of disorders with complex pathogenesis, including oxidative stress and microbial dysbiosis. The aim this study was to analyze the effect intracolonic administration plasma‐activated water (PAW) on status gut microbiota diversity in colon healthy mice IBD. Interestingly, PAW increased markers tissue, more pronounced IBD mice. Our results show that increases decreases it an inflamed gut. We conclude bidirectional depends underlying...

Microbial composition and activity were recorded during preparation of Pinot blanc (“Rulandské biele”) from one vineyard the years 2018, 2019, 2020. Both fungi bacteria are important for primary fermentation malto-lactic fermentation, therefore total DNA RNA isolated, genes 16S 28S rRNA amplified to determine both bacterial yeast profiles. Four phases wine production selected testing. We tested initial grape juice right after crushing grapes, must ~2-3 days post inoculation, actively...

Cronobacter is a ubiquitous Gram-negative bacterium linked with serious infections. In this report, we describe the characterization of phage Dev_CS701, which was isolated from wastewater. Related to phages belonging Straboviridae family and Pseudotevenvirus genus, such as vB_CsaM_IeB, Dev_CS701 contains 257 predicted protein-coding genes tRNA gene.

This study reports on a fatal case of captive great grey owl infected with the West Nile virus (WNV) in zoological garden Košice, eastern Slovakia (Central Europe). The tissue samples dead were used for isolation and genetic characterization. novel isolate is genetically closer to Hungarian, Greek, Bulgarian strains from central/southern European clade lineage 2 than previously isolated Slovakia. Interestingly, it carries NS3-249P, molecular virulence determinant associated higher...
