ABSTRACT Pulmonary vascular disease is not a single condition; rather it can accompany variety of pathologies that impact the pulmonary vasculature. Applying precision medicine strategies to better phenotype, diagnose, monitor, and treat increasingly possible with growing accessibility powerful clinical research tools. Nevertheless, challenges exist in implementing these tools optimal effect. The 2023 Grover Conference Series reviewed landscape summarize current state art provide...

Our lab reported the novel finding that stimulation of pulmonary endothelial cells with a beta-agonist and phosphodiesterase inhibitor induced enrichment cyclic adenosine monophosphate (cAMP) in EVs. cAMP protects vasculature by regulation permeability, vasoreactivity proliferation. Extracellular vesicles provide protected environment for circulating cAMP. Thus, we hypothesized administration cAMP, an EV, could deliver to improve outcomes hypertension. Briefly, male Sprague Dawley rats,...

The loss of ten-eleven translocation (TET2) methylcytosine dioxygenase expression contributes to the pathobiology pulmonary arterial hypertension (PAH). However, whether and activity other TETs DNA methyltransferases (DNMTs) are altered in PAH remains enigmatic. Therefore, our objective was determine DNMT (1, 3a, 3b) TET 2, 3) their total activity. We assessed enzymes leukocytes extracellular vesicles (EVs). Expression 3b), (2 leukocytes, EVs, from patients higher than healthy controls....

Abstract It is recently discovered that the cyclic nucleotide, adenosine monophosphate (cAMP) can be enriched in extracellular vesicles (EVs) isolated from endothelial cells. In current perspective a historical context for discovery of cAMP provided. The story through investigations addressing molecule's role pathway followed, which widespread mammalian physiology. mediates normal physiological conditions such as renin release, phosphate transport, etc., and participates pathological...

Second messenger signaling is required for cellular processes. We previously reported that extracellular vesicles (EVs) from stimulated cultured endothelial cells contain the biochemical second messenger, cAMP. In current study, we sought to determine whether cAMP-enriched EVs induce pathways in naïve recipient cells. Our results indicate increase cAMP content sufficient stimulate PKA activity. The implications of our work are represent a novel intercellular mechanism specifically cAMP, signaling.

Sex differences are recognized in pulmonary hypertension, however the progression of disease with regards to vascular lesion formation and circulating cytokines/chemokines is unknown. To determine whether formation, changes hemodynamics alterations chemokines/cytokines differ between male female. We used a progressive model PAH, SU/Hx analyzed cohorts female rats at timepoints suggested indicate worsening disease. Our analysis included echocardiograpy for hemodynamics, morphometry,...
