Purpose: Electronic cigarettes (e-cigs) are relatively new devices that allow the user to inhale a heated and aerosolized solution. At present, little is known about their health effects in human lung, particularly small airways (< 2 mm diameter), key site of airway obstruction destruction chronic obstructive pulmonary disease other acute lung conditions. The aim this study was investigate effect e-cigarettes on distal inflammation remodeling. Methods: We isolated primary epithelial cells...

The second messenger, cAMP, is highly compartmentalized to facilitate signaling specificity. Extracellular vesicles (EVs) are submicron, intact released from many cell types that can act as biomarkers or be involved in cell-to-cell communication. Although it well recognized EVs encapsulate functional proteins and RNAs/miRNAs, currently unclear whether cyclic nucleotides encapsulated within provide an additional messenger compartment. Using ultracentrifugation, were isolated the culture...

The loss of pulmonary endothelial cells in emphysema is associated with increased lung ceramide. Ceramide perturbations may cause adaptive alterations other bioactive sphingolipids, pathogenic implications. We previously reported a negative correlation between and circulating glycosphingolipids (GSLs). Glucosylceramide (GlcCer), the initial GSL synthesized from ceramide by GCS (GlcCer synthase), required for embryonic survival, but its role unknown.

Microparticles (MPs) are released constitutively and from activated cells. MPs play significant roles in vascular homeostasis, injury, as biomarkers. The unique glycocalyx on the membrane of cells has frequently been exploited to identify specific cell types, however yet be defined. Thus, we sought determine whether MPs, both during have a matching those parental type provide information MP origin. For these studies used rat pulmonary microvascular artery endothelium, smooth muscle, aortic...

Destruction of alveoli by apoptosis induced cigarette smoke (CS) is a major driver emphysema pathogenesis. However, when compared to cells isolated from non-smokers, primary human lung microvascular endothelial (HLMVECs) chronic smokers are more resilient exposed apoptosis-inducing ceramide. Whether this adaptation restores homeostasis unknown. To better understand the phenotype HLMVEC in smokers, we interrogated pro-survival pathway supported sphingosine-1-phosphate (S1P) signaling via S1P...

A frequently used end point of clinical outcomes in patients with pulmonary arterial hypertension (PAH) is the 6-minute walk distance. Furthermore, some data suggest that mild to moderate exercise as an intervention stable PAH beneficial. Some these questions have been recapitulated monocrotaline and hypoxia animal models hypertension. However, distance points not rigorously examined severe progressive Sugen 5416/hypoxia/normoxia (Su/Hx/Nx) model at each stage worsening disease. Our...

Abstract It is recently discovered that the cyclic nucleotide, adenosine monophosphate (cAMP) can be enriched in extracellular vesicles (EVs) isolated from endothelial cells. In current perspective a historical context for discovery of cAMP provided. The story through investigations addressing molecule's role pathway followed, which widespread mammalian physiology. mediates normal physiological conditions such as renin release, phosphate transport, etc., and participates pathological...

Second messenger signaling is required for cellular processes. We previously reported that extracellular vesicles (EVs) from stimulated cultured endothelial cells contain the biochemical second messenger, cAMP. In current study, we sought to determine whether cAMP-enriched EVs induce pathways in naïve recipient cells. Our results indicate increase cAMP content sufficient stimulate PKA activity. The implications of our work are represent a novel intercellular mechanism specifically cAMP, signaling.

Rationale Microparticles (MPs) are submicron intact vesicles released from many cell types constitutively or following activation. MPs play a role in cellular homeostasis, injury and act as biomarkers. These can encapsulate variety of functional proteins RNAs/miRNAs; however, little is known about whether they signaling molecules, such cyclic adenosine monophosphate (cAMP). Thus, we investigated the hypothesis that endothelial cells contain cAMP pulmonary microvascular (PMVEC) be stimulated...
