Viral haemorrhagic septicaemia (VHS) caused by the rhabdovirus VHSV is economically most important viral disease in European rainbow trout farming. Until 1989, this virus was mainly isolated from freshwater salmonids but last decade, it has also been an increasing number of free-living marine fish species. To study genetic evolution VHSV, entire G gene 74 isolates analysed. wild species caught Baltic Sea, Skagerrak, Kattegat, North and English Channel isolates, appeared to share a recent...
Deoxyribonucleic acid (DNA) vaccination is based on the administration of gene encoding vaccine antigen, rather than antigen itself. Subsequent expression by cells in vaccinated hosts triggers host immune system. Among many experimental DNA vaccines tested various animal species as well humans, against rhabdovirus diseases fish have given some most promising results. A single intramuscular (IM) injection microgram amounts induces rapid and long-lasting protection farmed salmonids...
Egtved virus, the rhabdovirus causing viral haemorrhagic septicaemia in rainbow trout, was analysed at antigen level with a future subunit vaccine mind. Three monoclonal antibodies to G protein were characterized respect neutralizing activity cell culture level, as well their ability protect trout fingerlings against virus infection following passive immunization. Two showed strong protective fish. Only one of these able neutralize infectivity vitro. Reduction disulphide bonds abolished...
With the aim of studying molecular mechanisms virulence and immunogenesis Egtved virus, monoclonal antibodies (MAbs) were produced against 4 dominant virus proteins (G, N, M, M2).The reactivity each MAb was determined by ELISA, immunoblotting, immunofluorescence plaque neutralization.Antibodies specific for proteins, as demonstrated gave characteristic reactions in ELISA well immunofluorescence.None MAbs able to neutralize vitro. When analysed using cell cultures fixed at different times...
The gene encoding the envelope glycoprotein of a recent Danish isolate salmonid rhabdovirus, viral haemorrhagic septicaemia virus (VHSV) has been cloned and sequenced at cDNA level. When compared with deduced sequence French VHSV, it was noted that there were 13 amino acid substitutions in virus. Amino homologies North American rhabdovirus (infectious haematopoietic necrosis virus) indicate high degree structural similarity between two fish glycoproteins. Results from partial enzymatic...
Although Spring Viraemia of Carp Virus (SVCV) can cause high mortalities in common carp, a commercial vaccine is not available for worldwide use. Here, we report DNA based on the expression SVCV glycoprotein (G) which, when injected muscle even at single low dose 0.1 µg DNA/g fish, confers up to 100% protection against subsequent bath challenge with SVCV. Importantly, best validate efficacy, also optimized reliable model closely mimicking natural infection, prolonged exposure carp 15°C....
Viral hemorrhagic septicemia (VHS) in rainbow trout Oncorhynchus mykiss is caused by VHS virus (VHSV), which belongs to the rhabdovirus family. Among different strategies for immunizing fish with a recombinant vaccine, genetic immunization has recently proven be highly effective. To further investigate potential protecting against DNA vaccination, experiments were conducted determine amount of plasmid needed induction protective immunity. The time onset immunity and duration protection...
