ABSTRACT We investigated the significance of endogenous reactive oxygen species (ROS) produced by fungi treated with miconazole. ROS production in Candida albicans was measured a real-time fluorogenic assay. The level increased miconazole at MIC (0.125 μg/ml) and enhanced further dose-dependent manner, fourfold increase detected when used 12.5 μg/ml. This completely inhibited pyrrolidinedithiocarbamate (PDTC), an antioxidant, 10 μM. In colony formation assay, decrease cell viability...
The aim of the present study was to assess contribution polymorphisms in breast cancer resistance protein/ATP-binding cassette transporter G2 (<i>BCRP/ABCG2</i>) gene placental expression from a new perspective, allelic imbalance. Polymorphisms were screened by polymerase chain reaction (PCR)-single-strand conformation polymorphism analysis followed sequencing with DNA extracted 100 placentas. To examine whether <i>BCRP</i> correlate BCRP expression, we determined mRNA and protein levels...
We examined whether survivin acts as a constitutive and inducible radioresistance factor in pancreatic cancer cells. Using quantitative TaqMan reverse transcription-polymerase chain reaction for mRNA five cell lines, we found an inverse relationship between expression radiosensitivity. PANC-1 cells, which had the highest levels, were most resistant to X-irradiation; MIAPaCa-2 showed least expression, sensitive X-irradiation. Our results suggested that could act To determine is enhanced by...
Induction of olfactomedin 4 (OLFM4 GW112/hGC‐1 ) in cancer cells was recently reported to have a novel antiapoptotic action via binding the potent apoptosis inducer GRIM‐19. We sought clarify undiscovered functions constitutively expressed OLFM4 cells. mRNA highly pancreatic tissues. In PANC‐1 cell cultures, expression especially elevated during early S phase cycle. Transduction small interfering RNA for decrease caused time‐dependent growth inhibition, with typical S‐phase arrest after 6...
In Japan, S-1 is used as adjuvant chemotherapy for pancreatic cancer. Neutropenia during reported to be an independent predictor of prolonged survival in patients with advanced gastric However, this unclear This study aimed examine the effect severe neutropenia caused by on cancer prognosis: overall (OS) and recurrence-free (RFS), potential duration from surgery administration OS RFS. single-center, retrospective, observational included who newly received after curative resection at Japanese...
We established the validity of a quantitative reverse transcription (RT)-PCR assay for RNA component human telomerase (hTR), using TaqMan fluorogenic detection system. Using this assay, we quantified hTR expression in two pancreatic cancer cell lines, ASPC-1 and MIAPaCa-2. Our results indicated that MIAPaCa-2 was 1.99-fold higher than cells. This RT-PCR appears to be useful determining amount clinical specimens.
Using gene-transduced pancreatic cancer cells, we examined whether survivin expression is directly involved in regulation of radiosensitivity. Ordinarily radiosensitive MIAPaCa-2 cells transduced with wild-type gene (MS cells) proliferated more rapidly than control vector. MS were significantly less vector-transduced cells. Radiation-induced activity caspase-3, but not caspase-7, was inhibited On the other hand, transduction a dominant-negative mutant into radioresistant PANC-1 augmented...
Abstract Background: Tumor necrosis factor-α (TNFα) is an important mediator of inflammatory and autoimmune diseases. Analysis its pathophysiologic roles has been difficult because low concentrations TNFα, including those in healthy controls, cannot be measured by existing methods. Methods: We developed a sensitive immuno-PCR assay for the detection TNFα human serum. The DNA label was generated PCR amplification using biotinylated primer bound with streptavidin to third antibody. sandwiched...
Recently, a number of nucleotide variants have been described in the multidrug resistance 1 (<i>MDR1/ABCB1</i>) gene; however, most studies focused on coding region. In present study, we identified promoter <i>MDR1</i> gene and evaluated their phenotypic consequences using reporter assay real-time polymerase chain reaction method. Ten allelic were detected region (approximately 2 kilobases), seven which newly identified. Certain mutations occurred simultaneously, total 10 haplotypes...
Overexpression of the olfactomedin 4 ( OLFM4 GW112,/hGC−1 ) gene was recently reported to inhibit various apoptotic pathways and promote proliferation cancer cells, suggesting that might serve as a diagnostic marker for human cancers. Therefore, we examined cancer‐specific overexpression. mRNA highly expressed in cancerous tissues obtained from colon, breast lung. Positivity mRNA, defined mean + 2 SD non‐cancerous colon tissues, observed 68 50% studied tissues. expression not detected lung...
The polyphenol (-)-epigallocatechin-3-gallate (EGCG) is a green tea constituent, which has been shown to inhibit cancer cell growth in vitro, vivo and epidemiological studies. In this study, we investigated its effects gastric lines. Five lines, the MKN-1, MKN-28, MKN-45, NUGC-3 TMK-1, were found be sensitive EGCG treatment. Of all lines tested, was most sensitive. treatment of cells induced apoptosis, confirmed by sub-G1 analysis, caspase-Glo assay Western blotting against cleaved PARP...
Background: Medication adherence has an important influence on health outcomes in patients with chronic diseases. However, few studies have been performed Japan to determine factors related medication non-adherence. Objective: The aim of this study was identify prescription non-adherence by investigating patient characteristics, all prescriptions, and prescriptions for oral antidiabetic drugs (OADs). Methods: A retrospective cross-sectional survey data about implementation dosing regimen at...
With most immunological reactions, tissue fibrosis, collagen overproduction caused by immune cytokines, is inevitably associated. Among the various heat shock protein 47 (HSP47) a procollagen-specific molecular chaperon and essential for secretion of procollagen from cells. Induction HSP47 TGF-beta has been previously reported in rat skeletal myoblasts mouse osteoblasts, but not human diploid fibroblasts. As IL-1beta, its effect on elucidated. In present study, using embryonic lung...
Survivin, a member of the inhibitor‐of‐apoptosis family, inhibits apoptosis by blocking caspase‐3 and ‐7 activation. Gastric cancer, which is among most intractable malignant tumors, known for resistance to various drugs, including cis ‐diamminedichloroplatinum (CDDP). Since this agent induces via activation, survivin may mediate drug resistance. We investigated messenger RNA (mRNA) expression in gastric cancers relationship between sensitivity CDDP. Expression gene was significantly...
Selective serotonin reuptake inhibitor (SSRI)-induced nausea can be severe enough to lead early treatment discontinuation. However, it is currently not possible predict the occurrence of before initiation SSRI treatment. In this study, we investigated effect genetic polymorphisms in 5-hydroxytryptamine type 2A, 3A, and 3B (5-HT3B) receptors, 5-HT transporter, CYP2D6 genes on incidence paroxetine-induced nausea. A consecutive series 72 Japanese patients with depressive or anxiety disorders...
3-Hydroxy-3-methylglutaryl coenzyme A reductase inhibitors (HRIs) are widely used to reduce serum cholesterol in patients with hypercholesterolemia. Previous studies have shown that HRIs can induce apoptosis colon cancer cells. In this study, we investigated the mechanisms underlying apoptosis-inducing effect of greater detail. The HRI lovastatin induced human cell line SW480 by blocking synthesis pathway. Immunoblot analysis antiapoptotic molecules, including survivin, XIAP, cIAP-1, cIAP-2,...
