ABSTRACT We investigated the significance of endogenous reactive oxygen species (ROS) produced by fungi treated with miconazole. ROS production in Candida albicans was measured a real-time fluorogenic assay. The level increased miconazole at MIC (0.125 μg/ml) and enhanced further dose-dependent manner, fourfold increase detected when used 12.5 μg/ml. This completely inhibited pyrrolidinedithiocarbamate (PDTC), an antioxidant, 10 μM. In colony formation assay, decrease cell viability...

The aim of the present study was to assess contribution polymorphisms in breast cancer resistance protein/ATP-binding cassette transporter G2 (<i>BCRP/ABCG2</i>) gene placental expression from a new perspective, allelic imbalance. Polymorphisms were screened by polymerase chain reaction (PCR)-single-strand conformation polymorphism analysis followed sequencing with DNA extracted 100 placentas. To examine whether <i>BCRP</i> correlate BCRP expression, we determined mRNA and protein levels...

We examined whether survivin acts as a constitutive and inducible radioresistance factor in pancreatic cancer cells. Using quantitative TaqMan reverse transcription-polymerase chain reaction for mRNA five cell lines, we found an inverse relationship between expression radiosensitivity. PANC-1 cells, which had the highest levels, were most resistant to X-irradiation; MIAPaCa-2 showed least expression, sensitive X-irradiation. Our results suggested that could act To determine is enhanced by...

Induction of olfactomedin 4 (OLFM4 GW112/hGC‐1 ) in cancer cells was recently reported to have a novel antiapoptotic action via binding the potent apoptosis inducer GRIM‐19. We sought clarify undiscovered functions constitutively expressed OLFM4 cells. mRNA highly pancreatic tissues. In PANC‐1 cell cultures, expression especially elevated during early S phase cycle. Transduction small interfering RNA for decrease caused time‐dependent growth inhibition, with typical S‐phase arrest after 6...

We established the validity of a quantitative reverse transcription (RT)-PCR assay for RNA component human telomerase (hTR), using TaqMan fluorogenic detection system. Using this assay, we quantified hTR expression in two pancreatic cancer cell lines, ASPC-1 and MIAPaCa-2. Our results indicated that MIAPaCa-2 was 1.99-fold higher than cells. This RT-PCR appears to be useful determining amount clinical specimens.

Using gene-transduced pancreatic cancer cells, we examined whether survivin expression is directly involved in regulation of radiosensitivity. Ordinarily radiosensitive MIAPaCa-2 cells transduced with wild-type gene (MS cells) proliferated more rapidly than control vector. MS were significantly less vector-transduced cells. Radiation-induced activity caspase-3, but not caspase-7, was inhibited On the other hand, transduction a dominant-negative mutant into radioresistant PANC-1 augmented...

Abstract Background: Tumor necrosis factor-α (TNFα) is an important mediator of inflammatory and autoimmune diseases. Analysis its pathophysiologic roles has been difficult because low concentrations TNFα, including those in healthy controls, cannot be measured by existing methods. Methods: We developed a sensitive immuno-PCR assay for the detection TNFα human serum. The DNA label was generated PCR amplification using biotinylated primer bound with streptavidin to third antibody. sandwiched...

Recently, a number of nucleotide variants have been described in the multidrug resistance 1 (<i>MDR1/ABCB1</i>) gene; however, most studies focused on coding region. In present study, we identified promoter <i>MDR1</i> gene and evaluated their phenotypic consequences using reporter assay real-time polymerase chain reaction method. Ten allelic were detected region (approximately 2 kilobases), seven which newly identified. Certain mutations occurred simultaneously, total 10 haplotypes...
