Researchers working with fission yeast conduct protein extraction widely and frequently, but this includes the handling of glass beads, hence is laborious cumbersome, especially when dealing a large number samples. Here we describe rapid reliable method for preparing extract from yeast, one which applicable to routine western blotting.

Dynamic microtubule plus-ends interact with various intracellular target regions such as the cell cortex and kinetochore. Two conserved families of plus-end-tracking proteins, XMAP215, ch-TOG or CKAP5 family end-binding 1 (EB1, also known MAPRE1) family, play pivotal roles in regulating dynamics. Here, we study functional interplay between fission yeast Dis1, a member XMAP215/TOG Mal3, an EB1 protein. Using vitro microscopy assay, find that purified Dis1 autonomously tracks growing ends is...

Gap-repair cloning for plasmid construction in budding yeast is very effective and often used. In contrast, the same method not widely used fission yeast, because of a shortage information on it. Here we describe simple gap-repair using short tracts flanking homology. By this method, combined concentrated DNA fragments with (20 bp) homology marker gene or pre-existing module. addition, found that can be applied to one-step multiple construct fusion gene.

The histone H3-H4 chaperone Asf1 is involved in chromatin assembly (or disassembly), exchange, regulation of transcription, and silencing several organisms. To investigate the essential functions Schizosaccharomyces pombe, asf1-ts mutants were constructed by random mutagenesis using PCR. One mutant (asf1-33(ts)) was mated with 77 different kinase genes to identify synthetic lethal combinations. asf1-33 required DNA damage checkpoint factors Chk1 Rad3 for its survival at restrictive...

A considerable portion of agricultural land in central-east Japan has been contaminated by radioactive material, particularly Cs, due to the industrial accident at Fukushima Daiichi nuclear power plant. Understanding mechanism absorption, translocation and accumulation Cs+ plants will greatly assist developing approaches help reduce contamination products. At present, however, little is known regarding transporters rice. transporter-enriched yeast expression library was constructed screened...

Polypeptone is widely excluded from Schizosaccharomyces pombe growth medium. However, the reasons why polypeptone should be avoided have not been documented. dramatically induced cell lysis in ura4 deletion mutant when cells approached stationary phase, and this phenotype was suppressed by supplementation of uracil. To determine specificity phenotype, we created mutants other genes involved de novo biosynthesis uridine monophosphate (ura1, ura2, ura3, ura5). Cell observed these gene mutants....

High-fidelity chromosome segregation relies on proper microtubule regulation. Kinesin-8 has been shown to destabilise microtubules reduce metaphase spindle length and movements in multiple species. XMAP215/chTOG polymerases catalyse growth for assembly, elongation kinetochore-microtubule attachment. Understanding of their biochemical activity advanced, but little work directly addresses the functionality interplay these conserved factors. We utilised synthetic lethality fission yeast...

An H3/H4 histone chaperone, Asf1, plays an essential role in maintaining genomic stability many species, including fission yeast. The Ndc80 complex is a conserved outer kinetochore protein consisting of (Hec1), Nuf2, Spc24 and Spc25. This comprises major, if not the sole, platform with which plus ends spindle microtubules directly interact. In fission yeast, several studies indicate that multiple microtubule-associated proteins including Dis1/chTOG microtubule polymerase Mal3/EB1 plus-end tracking or indirectly bind Ndc80, thereby ensuring stable kinetochore-microtubule attachment. However, purification from this yeast...

Schizosaccharomyces pombe Δura4 cells lyse when grown on YPD medium. A S. non-essential gene deletion library was screened to determine suppressors of the lysis phenotype. Deletion pub1 gene, which encoded E3 ubiquitin ligase, strongly suppressed cell in cells. The Δpub1 displayed high sensitivity 5-fluorouracil, a toxic analog uracil, and this by fur4, uracil transporter. Fur4 localized primarily Golgi apparatus vacuoles wild-type cells, but localization predominantly at plasma membrane...
