Understanding the composition and function of vaginal microbiome is crucial for reproductive overall health. Here we established Isala citizen-science project to analyse microbiomes 3,345 women in Belgium (18-98 years) through self-sampling, 16S amplicon sequencing extensive questionnaires. The was strongly tied age, childbirth menstrual cycle phase. Lactobacillus species dominated 78% samples. Specific bacterial taxa also showed co-occur modules based on network correlation analysis....

Riboflavin-producing lactic acid bacteria represent a promising and cost-effective strategy for food biofortification, but production levels are typically insufficient to support daily human requirements. In this study, we describe the novel isolate Limosilactobacillus reuteri AMBV339 as strong biofortification candidate. This strain shows high natural riboflavin (vitamin B2) overproduction of 18.36 μg/ml, biomass up 6 × 1010 colony-forming units/ml (in typical range model lactobacilli),...

Despite its importance in health, the female skin microbiome remains understudied. In this study we explored microbial dispersal across intimate body sites, including vagina, groins, breast and mouth. Microbial similarity correlated with physical proximity, suggesting influenced by hygiene or sexual activity. Notably, lactobacilli were unexpectedly abundant on skin. These findings highlight need for research into dynamics their implications women's health.

This proof-of-concept study compares first-void urine (FVU) and cervicovaginal brushes to vaginal swabs for microbiome profiling in ten women. FVU provided comparable results most cases, though storage conditions influenced microbial composition, especially FVU. Despite limitations sample size diversity participants, findings support these methods' potential self-sampling women's health research, emphasizing the need optimized techniques improve profiling.

Few validated protocols are available for large-scale collection, storage, and analysis of microbiome samples from the vagina, skin, mouth. To prepare a study on female by remote self-sampling, we investigated impact sample host DNA depletion profiling. Vaginal, saliva were analyzed using 16S rRNA gene amplicon metagenomic shotgun sequencing, qPCR. Of two tested storage buffers, eNAT buffer could keep microbial composition stable during various conditions. All three DNA-depletion approaches...

<title>Abstract</title> Predicting recent intercourse is crucial in forensic casework on sexual assaults. In this work, we assessed whether can be predicted based the vaginal microbiome and compared it to gold standard method of semen detection. Using a prediction model 3,043 women, was with 71% accuracy balanced cross-validation machine learning setting. Next, validated longitudinal intervention study tested assault cases. The developed predictor could accurately establish 82% Yet,...

Abstract The vaginal microbiome is crucial for women’s health and reproduction, but its ecology determinants in the general population are still unclear. This lack of a reference framework hampers much-needed innovations diagnostics therapeutics. Here, we remotely mapped 3,345 women Western Europe via citizen-science approach. More than 75% samples were dominated by Lactobacillus taxa, not discrete community state types. Compositional correlation network analysis validated with public data...

Introduction The pathogenesis of Attention-Deficit Hyperactivity Disorder (ADHD) is thought to be multifactorial, with a potential role for the bidirectional communication between gut microbiome and brain development function. Since “golden-standard” medication therapy methylphenidate (MPH) linked multiple adverse effects, there need alternative treatment options such as dietary polyphenols. These secondary plant metabolites exert antioxidant anti-inflammatory but much less known about their...

A novel strain of the genus Lactobacillus, named AMBV1719T, was isolated from vagina a healthy participant in our large-scale citizen science project on female microbiome, Isala. Phylogenetic analysis showed that 16S rRNA gene AMBV1719T is most similar to Lactobacillus taiwanensis with sequence similarity 99.873 %. However, genome-wide comparison using average nucleotide identity (ANI) revealed isolate highest ANI paragasseri JCM 5343T, value only 88.17 This low closely related strains known...

Abstract Background Due to the lowering of circulating estrogens symptoms genitourinary syndrome menopause (GSM) may arise with premenopausal breast cancer patients who become menopausal due treatment. Clinical signs GSM are i.a. vaginal dryness, dyspareunia, loss libido, dysfunction arousal and/or orgasm: this negatively impact health, sexual activity and quality life. The composition function microbiome also change changes gonadal hormones during transition. This contribute discomfort...
