How environmental factors combine with genetic risk at the molecular level to promote complex trait diseases such as multiple sclerosis (MS) is largely unknown. In mice, N-glycan branching by Golgi enzymes Mgat1 and/or Mgat5 prevents T cell hyperactivity, cytotoxic T-lymphocyte antigen 4 (CTLA-4) endocytosis, spontaneous inflammatory demyelination and neurodegeneration, latter pathologies characteristic of MS. Here we show that MS modulators converge alter N-glycosylation CTLA-4 surface...

Essential biological systems employ self-correcting mechanisms to maintain cellular homeostasis. Mammalian cell function is dynamically regulated by the interaction of surface galectins with branched N-glycans. Here we report that N-glycan branching deficiency triggers Golgi generate bioequivalent N-glycans preserve galectin-glycoprotein interactions and Galectins bind N-acetyllactosamine (LacNAc) units within initiated from UDP-GlcNAc medial-Golgi enzymes as well trans-Golgi poly-LacNAc...

Summary Objective Although the link between vigabatrin ( VGB ) and retinotoxicity is well known, little attention has been focused on risk of ‐associated brain abnormalities magnetic resonance imaging MRI VABAM ), namely reversible—and largely asymptomatic—signal changes in thalami, basal ganglia, brainstem tegmentum, cerebellar nuclei. Using a large infantile spasms cohort, we set out to identify predictors these phenomena. Methods Children with were retrospectively identified. Brain...

Epidemiologic studies correlate low vitamin C intake with bone loss. The genetic deletion of enzymes involved in de novo synthesis mice, likewise, causes severe osteoporosis. However, very few have evaluated a protective role this dietary supplement on the skeleton. Here, we show that ingestion prevents low-turnover loss following ovariectomy mice. We prevention areal mineral density and micro-CT parameters results from stimulation formation, demonstrable vivo by histomorphometry, marker...

Abstract N-glycan branching is a potent and multifaceted negative regulator of proinflammatory T cell B function. By promoting multivalent galectin–glycoprotein lattice formation at the surface, regulates clustering and/or endocytosis TCR complex (TCR+CD4/CD8), CD45, CD25, BCR, TLR2 TLR4 to inhibit activation/proliferation TH1 TH17 over TH2 induced regulatory responses. In addition, promotes surface retention growth inhibitory receptor CTLA-4. However, role in regulating levels other...

Abstract Micropipette tip (tip) pipetting accuracy and precision are functions of manufacture consistency, morphological variation, retained inner wall particulate matter. Irregular surfaces matter cause inaccuracy. Washed unwashed tips from seven manufacturers were compared using weight by difference (six), microscopic Luxol Fast Blue staining (seven), matrix‐assisted laser desorption/onization time‐of‐flight (MALDI‐TOF) mass spectrometry (six). Photomicrographs revealed irregularity...

Abstract Aging is associated with a decline in immune function, increasing the frequency and severity of infection elderly. We have previously shown that Asparagine (N) – linked protein glycosylation critical negative regulator T cell immunity both mice humans. Here we report N-glycan branching cells increases age, leading to hypo-activity increased susceptibility infection. Reducing rejuvenates activation, proliferation pro-inflammatory TH17 over anti-inflammatory Treg differentiation aged...

A 54-year old female with longstanding metastatic breast cancer was referred for management of choroidal metastases. She first treated external beam radiotherapy and experienced some response before later suffering progression her eye disease. We then using full fluence Photodynamic therapy (PDT) Verteporfin, which resulted in regression lesions until she passed away due to other illnesses. This is the documented successful application PDT metastasis from a primary refractory radiotherapy.
