We provide the initial performance evaluation of a 21 Tesla Fourier transform ion cyclotron resonance mass spectrometer operating at Environmental Molecular Sciences Laboratory Pacific Northwest National Laboratory. The constructed for 21T system employs commercial dual linear trap coupled to FTICR designed and built in-house. Performance gains from moving higher magnetic field strength are exemplified by measurement peptide isotopic fine structure, complex natural organic matter mixtures,...
Specimens of Scomberoides tala (Cuvier, 1832) were collected from southwestern Taiwan in the northwestern Pacific Ocean. While this species has been documented other regions Pacific, our specimens mark its first recorded occurrence Taiwan, bridging a previously identified distribution gap region. Additionally, we suggest that may be shifting northward, potentially driven by global warming, as northern records reported rather recently.
Cisplatin is a potent anticancer drug, which functions by cross-linking adjacent DNA guanine residues. However within 1 day of injection, 65–98% the platinum in blood plasma protein-bound. It generally accepted that cisplatin binds to methionine and histidine residues, but what often underappreciated from has 2+ charge can form up four bonds. Thus, it potential function as cross-linker. In this report, ability demonstrated Fourier transform ion cyclotron resonance (FTICR) mass spectrometry...
Calmodulin (CaM) is a highly conserved, ubiquitous, calcium-binding protein; it binds to and regulates many different protein targets, thereby functioning as calcium sensor signal transducer. CaM contains 9 methionine (Met), 1 histidine (His), 17 aspartic acid (Asp), 23 glutamine (Glu) residues, all of which can potentially react with platinum compounds; thus, one-third the sequence possible binding target anticancer drugs, represents major challenge for identification specific modification...
The short and long isoforms of the dopamine D2 receptor (D2S D2L respectively) are highly expressed in striatum. Functional receptors activate an intracellular signalling pathway that includes a cAMP-independent route involving Akt/GSK3 (glycogen synthase kinase 3). To investigate response to seldom-studied D2S receptor, we established rat receptor-expressing cell line [HEK (human embryonic kidney)-293/rD2S]. We found HEK-293/rD2S cells, D2/D3 agonists bromocriptine quinpirole significantly...
A novel single-transistor transimpedance preamplifier has been introduced for improving performance in Fourier-transform ion cyclotron resonance (FT-ICR) mass spectrometry. low noise junction field-effect transistor (JFET), BF862, is used as the main amplification stage of this trans-impedance preamplifier, and a T-shaped feedback network both gate biasing solutions. The T studied using an operational amplifier (Op Amp), AD8099. Such system allows ∼100-fold less resistance at given...
Our previous study has shown that tunicamycin irreversibly downregulates the expression of GABAAR and causes cell death in cultured brain neurons by biochemical light microscopic methods. In this study, we examined mechanisms underlying degeneration mainly employing electron analysis. Cultured derived from embryonic chicken brains were incubated with 5 μg/ml (TM) for 24 h, followed continual incubation or removal TM additional 3 h h. Neurons treated showed dilated rough endoplasmic reticulum...
The nature of the ion signal from a 12-T Fourier-transform cyclotron resonance mass spectrometer and electronic noise were studied to further understand detection limit. At minimal cost, new transimpedance preamplifier was designed, computer simulated, built, tested. design pushes signal-to-noise performance at room temperature limit, because its enhanced tolerance capacitance device, lower intrinsic noise, larger flat mid-band gain (input current spectral density around 1 pA/√Hz when is...
Two specimens representing the first known adults of Lestidiopsindopacificus (Ege, 1953) are reported and described from Taiwan, validity generic assignment this species confirmed. The origin pelvic fin directly below dorsal-fin base shows that L.indopacificus belongs to L.mirabilis complex. It can be separated its congeners by position nostrils above posterior end maxilla, light body color with unevenly distributed melanophores in adults, a distinct combination meristic values other...
The gibberfish Gibberichthyslatifrons is recorded for the first time from southwestern Taiwan, northern South China Sea, based on collection of seven adults and subadults Dong-gang fish market, Pingtung. These specimens represent northernmost extent adult range species, they fill a distributional gap in western Pacific Ocean. Our findings suggest that population species has become recently established region. Detailed descriptions, otolith morphology, fresh coloration are provided with...
The high mass accuracy and resolving power of Fourier transform ion cyclotron resonance spectrometers (FT-ICR MS) make them ideal detectors for spectrometry imaging (MSI), promising to provide unmatched molecular resolution capabilities. intrinsic low tolerance FT-ICR MS RF interference, however, along with typically vertical positioning the sample, MSI acquisition speed requirements present numerous engineering challenges in creating robotics capable achieving spatial match. This work...
13-Acetoxysarcocrassolide (13-AC) is a marine cembranoid derived from the aquaculture soft coral of Lobophytum crassum. The cytotoxic effect 13-AC against leukemia cells was previously reported but its mechanism action still unexplored. In current study, we showed that induced apoptosis human acute lymphoblastic Molt4 cells, as evidenced by cleavage PARP and caspases, phosphatidylserine externalization, well disruption mitochondrial membrane potential. use N-acetylcysteine (NAC), reactive...
Mechanisms underlying the pathogenesis of psychotic disorders were explored by monitoring expression GABAergic neurons in an animal model. Male rats postnatal days 21 and 60 intraperitoneally injected with amphetamine (Amph), 5 mgkg, or saline three times daily for 6 d. After 1-d 14-d withdrawal from Amph, they challenged on day 8 (W1d) (W14d) a single same dosage then perfused. Immunostaining brain sections using anti-glutamic acid decarboxylase (GAD67) antiserum revealed that Amph...
The significance of N-linked glycosylation and oligosaccharide processing was examined for the expression γ-aminobutyric acidA receptor (GABAAR) in cultured neurons derived from chick embryo brains. Incubation cultures with 5 μg/ml tunicamycin 24 h blocked binding 3H-flunitrazepam 3H-muscimol, probes benzodiazepine GABA sites on receptor, by about 20% 28%, respectively. loss ligand due to a reduction number no significant changes affinity. Light microscopic immunocytochemistry also revealed...
Phillyrin is isolated from the fruit of Forsythia suspensa, and exhibits multiple pharmacological effects, including anti-tumor, anti-inflammation, anti-oxidation activities. Here, we investigated whether phillyrin could alleviate airway hyperresponsiveness (AHR) eosinophil infiltration in lungs asthmatic mice, mitigate inflammatory responses tracheal epithelial BEAS-2B cells. IL-4/TNF-α-stimulated cells were treated with various doses. Female BALB/c mice sensitised challenged ovalbumin...

A transimpedance amplifier is a common preamplifier solution for capacitive charge/current detector, and widely used in optical communication. The concept ideal mass spectrometry (MS) system, particular the Fourier-Transform Ion Cyclotron Resonance (FT-ICR) MS, MS with highest resolution accuracy. To push performance toward prospective goal of single-ion detection, novel T Feedback Network was designed tested two preamplifiers. At given transimpedance, can reduce feedback resistance by ~100...
