Abstract The adaptation of CRISPR/SpCas9 technology to mammalian cell lines is transforming the study human functional genomics. Pooled libraries CRISPR guide RNAs (gRNAs) targeting protein-coding genes and encoded in viral vectors have been used systematically create gene knockouts a variety cancer immortalized lines, an effort identify whether these cause cellular fitness defects. Previous work has shown that screens are more sensitive specific than pooled-library shRNA similar assays, but...

The capacity of photosynthetic microorganisms to fix carbon dioxide into biomass positions them as promising cell factories for sustainable biomanufacturing. However, limitations in screening throughput hinder the identification enzymes, strains, and growth conditions needed realize this potential. Here we present a microplate-based high-throughput cultivation system that can be integrated existing automation infrastructure supports both prokaryotic eukaryotic microorganisms. We validate by...

Abstract Background Recent studies have uncovered the near-ubiquitous presence of microbes in solid tumors diverse origins. Previous literature has shown impact specific bacterial species on progression cancer. We propose that local microbial dysbiosis enables certain cancer phenotypes through provisioning essential metabolites directly to tumor cells. Methods 16S rDNA sequencing 75 patient lung samples revealed microbiome specifically enriched for bacteria capable producing methionine....

An embedding method requiring only 2 h to complete and giving excellent ultrastructural preservation has been used for the rapid detection of viruses in tissue cultures. The also applied successfully mammalian tissue. It provides a technique identifying isolated cultures, screening cultures adventitious agents, examining biopsies viruses.

Despite their recognized potential, current applications of cyanobacteria as microbial cell factories remain in early stages development. This is partly due to the fact that engineered strains are often difficult grow at scale. technical challenge contrasts with dense and highly productive populations thriving many natural environments. It has been proposed selection pre-adapted for growth industrial photobioreactors could enable more cultivation outcomes. Here, we described initial...

Abstract Monitoring population dynamics in co-culture is necessary engineering microbial consortia involved distributed metabolic processes or biosensing applications. However, it remains difficult to measure strain-specific growth high-throughput formats. This especially vexing plate-based functional screens leveraging whole-cell biosensors detect specific signals. Here we develop an experimental system and model the relationship between fluorescence cell abundance, combining...

Here, we report metagenome-assembled genomes for "Candidatus Phormidium sp. strain AB48" and three cooccurring microorganisms from a biofilm-forming industrial photobioreactor environment, using the PacBio sequencing platform. Several mobile genetic elements, including double-stranded DNA phage plasmids, were also recovered, with potential to mediate gene transfer within biofilm community.

We report the genome of Phormidium yuhuli AB48, which includes a circular chromosome and plasmid (4,747,469 bp 51,599 bp, respectively). This is currently only closed reference an isolate genus, based on Genome Taxonomy Database (GTDB), providing potential model system for sustainable biotechnology innovation.
