Abstract A number of scientific reports published to date contain data on endogenous levels various phytohormones in potato ( Solanum tuberosum L.) but a complete cytokinin profile tissues, that would include all particular molecular forms cytokinin, has still been missing. In this work, analytically detectable isoprenoid cytokinins, as well the auxin indole-3-acetic acid (IAA), and abscisic (ABA) have determined shoots roots 30 day old vitro grown (cv. Désirée). The results presented here...

Hairy root cultures of Hypericum perforatum were obtained following inoculation aseptically germinated seedlings with A. rhizogenes strain A4M70GUS. Effect sucrose on the growth and biomass production hairy was investigated. spontaneously regenerated shoots buds from which a number shoot culture clones established. Transformed exhibited good multiplication, elongation rooting hormone-free woody plant medium. Plants roots similar in appearance to normal, nontransformed plants.

Efficient protocols, safe from somaclonal variation, were developed for regeneration of Iris sibirica plants via organogenesis and somatic embryogenesis leaf-base explants cultivated on Murashige Skoog media supplemented with thidiazuron (TDZ, 1.0 mg/l) or 2,4-dichlorophenoxyacetic acid (2,4-D, mg/l). The morphogenic response callus formation efficiency differed significantly between 2,4-D (80.9%) TDZ (67%) morphogenesis induction treatments. induced only organogenic calli, while calli...

Abstract Colorado potato beetle (CPB; Leptinotarsa decemlineata Say, Coleoptera: Chrysomelidae) has shown a remarkable adaptability to variety of control measures. Although oryzacystatin I and II (OCI OCII) have potential in controlling pests that use cysteine proteinases for food digestion, expression single OC gene exhibited minimal or no effect on CPB fitness traits. The aim this study was examine the coexpressed OCI OCII ( Solanum tuberosum L.) cultivars Desiree, Dragačevka Jelica...

Allelopathic plants exploit their chemical 'weapons' to prevail over the competition, suppress neighboring and consequently use available resources more efficiently. However, investigation of plant allelopathic interactions in rhizosphere is difficult perform because its high complexity due biotic abiotic factors. Thus, autonomous, aseptic root cultures apple (Malus × domestica Borkh.) could facilitate studies. We report on successful genetic transformation cultivars Melrose, Golden...

Numerous biotechnological applications require a fast and efficient clonal propagation of whole plants under controlled laboratory conditions. For most plant species, the de novo regeneration shoots from cuttings various organs can be obtained on nutrient media supplemented with hormones, auxin cytokinin. While is needed during early stages process that include establishment pluripotent primordia subsequent acquisition organogenic competence, cytokinin-supplemented are required to induce...

Cross-talk between phytohormones and sugars is intensely involved in plant metabolism, growth regeneration. We documented alterations cytokinin (CK) homeostasis four developmental stages during de novo shoot organogenesis (DNSO) of kohlrabi (Brassica oleracea var. gongylodes cv. Vienna Purple) seedlings induced by exogenous CKs, trans-zeatin (transZ) thidiazuron (TDZ), added together with elevated sucrose concentration (6% 9%). Significant impact CK treatment their interaction was recorded...

Agrobacterium rhizogenes A4M70GUS-mediated transformation of Savoy cabbage (Brassica oleracea L. var. sabauda) and two local lines (B. capitata) was obtained using hypocotyl cotyledon explants. The percentage explants which formed roots very high in all genotypes: 92.3 % Gg-1, 64.4 P22I5, 87.2 P34I5. Spontaneous shoot regeneration excised root cultures grown on the hormone-free medium occurred three genotypes. In P22I5 P34I5 higher (9.3 2.6 respectively) than Gg-1 (1.3 %). Transgenic nature...

Non-transformed and transformed embryogenic cultures of alfalfa (Medicago sativa L. cv. Zaječarska 83), long-term maintained on growth regulator-free medium, were histologically analyzed. In all examined cultures, somatic embryos at various stages development observed secondary formed in the cotyledonary, hypocotylary radicular region primary embryos. Detailed histological analysis torpedo shape embryo revealed that arose directly from single epidermal cells axis after an unequal periclinal...

An efficient in vitro shoot regeneration method from leaf explants of apple cultivars Golden Delicious and Melrose by optimization medium, explant type orientation, dark pre-treatment, gelling agent is presented. Murashige Skoog’s medium containing 22 μM thidiazuron (TDZ) 1.5 indole-3-butyric acid (IBA) (M2 medium) was superior for as well subsequent multiplication both cultivars, providing frequency 95% or higher the best combination with other factors.
