Significance Transfer of sperm-producing stem cells isolated from a donor male into testes recipient has important applications for preserving germplasm and as conduit widespread dissemination desirable genetics in livestock production. A key aspect is surrogate males that lack endogenous germline but are otherwise physiologically normal. Here we demonstrate mice, pigs, goats rendered genetically sterile by CRISPR-Cas9 editing the NANOS2 gene support donor-derived spermatogenesis following...
Abstract Gene editing technologies, such as CRISPR-Cas9, have important applications in mammalian embryos for generating novel animal models biomedical research and lines of livestock with enhanced production traits. However, the lack methods efficient introduction gene reagents into zygotes various species need surgical embryo transfer mice been technical barriers widespread use. Here, we described methodologies that overcome these limitations mice, cattle, pigs. Using mutation Nanos2 a...
Abstract Cotranslational protein folding depends on general chaperones that engage highly diverse nascent chains at the ribosomes. Here we discover a dedicated ribosome-associated chaperone, Chp1, rewires cotranslational machinery to assist in challenging biogenesis of abundantly expressed eukaryotic translation elongation factor 1A (eEF1A). Our results indicate during eEF1A synthesis, Chp1 is recruited ribosome with help polypeptide-associated complex (NAC), where it safeguards biogenesis....
A 2-year-old Large White research gilt was presented to the Comparative Theriogenology service at WSU for infertility. She produced from transfer of genetically modified embryos obtained by in vitro fertilization (IVF) that had been treated with CRISPR-Cas9 reagents mutate NANOS2 gene. Since birth, showed a so-called “skyhook” vulva and abnormal estrous cycles. artificially inseminated multiple times, starting 5 months age, semen proven boar, but never conceived. On...

Heat Shock Factor 1 (Hsf1) in yeast drives the basal transcription of key proteostasis factors and its activity is induced as part core heat shock response. Exploring Hsf1 specific functions has been challenging due to essential nature HSF1 gene extensive overlap target promoters with environmental stress response (ESR) Msn2 Msn4 (Msn2/4). In this study, we constructed a viable hsf1∆ strain by replacing open reading frame genes that constitutively express Hsp40, Hsp70, Hsp90 from...
Heat shock factor 1 (HSF1) responds to stress mount the heat response (HSR), a conserved transcriptional program that allows cells maintain proteostasis by upregulating proteins (HSPs). The homeostatic regulation of HSF1 plays key role in human physiology and health but its mechanism has remained difficult pinpoint. Recent work budding yeast model implicated stress-inducible chaperones HSP70 family as direct negative regulators activity. Here, we have investigated latency control activation...
Male reproductive capacity is a critical component of cattle production and the majority genetic gain made via selective utilization gametes from desirable sires. Thus, strategies that enhance sperm increase availability elite genetics for use in improving characteristics populations on worldwide scale. In all mammals, amount produced strongly correlated to number Sertoli cells testes. Studies with rodents showed size cell population set during prepubertal development signaling thyroid...
Llamas and alpacas are important production animals in South America, with increasing interest other parts of the world. Poor reproductive efficiency combined several unique anatomical physiological features offer challenges diagnosis treatment infertility camelids. This review presents an approach to clinical investigation common causes subfertility male female. The selection males for breeding should be made based on complete evaluation eliminate congenital possibly hereditary disorders....
Abstract Heat shock protein 70 kDa (HSP70) is a major family in the cell protections against stress-induced denaturation and aggregation folding of nascent proteins. It highly conserved that can be found most organisms strongly connected to several intracellular pathways such as refolding, degradation regulation, protection intense stress. Cellular delivery HSP70 would high impact for clarification its role these cellular processes. PepFect14 cell-penetrating peptide known able mediate...

A 1-year-old male intact Miniature Schnauzer mix was presented for chronic intermittent hematuria. Abdominal ultrasonography revealed a large, fluid-filled cystic structure extending cranially and dorsally to the prostate. Computed tomography scan images that cavity resembled uterus, with both horns entering scrotum through inguinal canal adjacent testes. On cytogenetic analysis, dog found have homozygote mutation on AMHRII consistent persistent Müllerian duct syndrome (PMDS)....
Summary Uterine adenocarcinoma was diagnosed in a 16‐year‐old Quarter Horse mare that presented for fever, stranguria, anorexia and history of weight loss. A mass at the level cervix uterine body identified by transrectal palpation. Biopsy revealed an antemortem diagnosis adenocarcinoma. Thoracic ultrasonography multiple areas consolidation lung, comet tail artefact along entire pleural surface. Post‐mortem examination confirmed endometrial with metastasis to urinary bladder, ureters, lungs,...
HealthCenter Livestock Services in July 2023 to evaluate unilateral scrotal swelling. The bull was first noticed being ridden by other yearling bulls the pasture 1 day before presentation, which prompted owner notice is housed on a 160-acre with 75 and only animal affected. No history of prior medical conditions reported, he vaccinated against clostridial diseases. On physical examination, bright, alert, responsive had slightly increased heart rate (88 beats/min; reference, 50 80 beats/min),...
A 4-year German Shorthair Pointer pregnant dog was presented for severe tachypnea and depression. She had 1 natural breeding 52 days ago. Diaphragmatic hernia with intrathoracic displacement of the uterus diagnosed via radiography ultrasonography. Emergency herniorrhaphy elected owner requested to save litter if possible. Right uterine horn tip, spleen, part liver, pancreas, greater omentum were in thorax. Intraoperative transuterine ultrasonography confirmed live fetuses pregnancy permitted...
Alpacas have a high incidence of congenital reproductive tract abnormalities, including ovarian hypoplasia/dysgenesis. Diagnosis this condition is often challenging. The present study describes the clinical, ultrasonographic, and histologic features hypoplasia/dysgenesis syndrome in 5 female alpacas. Additionally, serum AMH levels were compared between alpacas diagnosed with group reproductively sound females ( n = 11). was suspected based on presence an infantile uterus lack ovaries by...
Development of bovine estrus synchronization protocols started in the 1970's.The aim these has changed progressively from synchronizing to ovulation with main goal performing insemination without heat detection (Timed artificial insemination). The present paper discusses physiology behind development and reviews used nowadays dairy herd reproductive management.
Earlier, we validated an anti-Müllerian hormone (AMH) enzyme-linked immunosorbent assay kit for male alpacas. First, wecompared the validation data with another kit. There was a high correlation (R2 = 0.94) between these 2 kits. Second, used thelatter to determine serum AMH concentrations during follicular and luteal phases of reproductive cycle in female alpacas. There were no differences (p 0.39) alpacas (n 11) peak phases(mean ± SEM, 1.33 0.35 versus 1.18 0.34 ng/ml, respectively). Third,...
