Trypanosoma cruzi, the parasite causing Chagas disease, is a digenetic flagellated protist that infects mammals (including humans) and reduviid insect vectors. Therefore, T. cruzi must colonize different niches in order to complete its life cycle both hosts. This fact determines need of adaptations face challenging environmental cues. The primary challenge, particularly stages, poor nutrient availability. In this regard, it well known has flexible metabolism able rapidly switch from...

Background: Considering the fact that many nitrogenated aromatic derivatives such as pyrazoles, hydrazones, pyrazolo[1,5-a]-pyrimidines and pyrazolo [3,4-b]-pyridine have shown antiparasitic properties, current study was carried out to analyze in vitro antileishmanial antitrypanosomal potential of some derivatives, synthesize from 6-substituted-3-formylcromone. Method: The pyrazoles (4a-c), hydrazones (5a-c), (6a, 6b) (7) were prepared by condensation reaction between 6-substituted...

ATP hydrolysis is required for the synthesis, transport and polymerization of monomers macromolecules as well assembly latter into cellular structures. Other processes not directly related to synthesis biomass, such maintenance membrane potential shape, also require ATP. The unicellular flagellated parasite Trypanosoma brucei has a complex digenetic life cycle. primary energy source this in its bloodstream form (BSF) glucose, which abundant host’s bloodstream. The protozoan parasite Leishmania causes a variety of sicknesses with different clinical manifestations known as leishmaniasis. chemotherapy currently in use is not adequate because their side effects, resistance occurrence, and recurrences. Investigations looking for new targets or active molecules focus mainly on the disruption specific pathways. In this sense, ergosterol biosynthesis one most attractive it does occur mammals. Here, we report synthesis ergosterone coupled characterization...

Among the scarce validated drug targets against Chagas disease (CD), caused by Trypanosoma cruzi, parasite's nucleoside salvage system has recently attracted considerable attention. Although trypanocidal activity of tubercidin (7-deazapurine) long been known, identification a class 7-substituted analogs with potent in vitro and vivo much-enhanced selectivity made among most promising lead compounds CD. Here, we investigate identified TcrNT2 transporter its potential role antimetabolite...

Abstract13,14-Dihydroxy-8,11,13-podocarpatrien-7-one (1) and a series of ring C aromatic diterpene derivatives were synthesised from (+)-manool (4) evaluated for their cytotoxic, leishmanicidal trypanocidal activities. Our results indicated that compound 1 other podocarpane-type intermediates are cytotoxic. Cleavage C6–C7 bond 7 improved cytotoxic activity, indicating that, in particular, the 6,7-seco-podocarpane-type 20 might serve as lead further development.Keywords::...

Chagas disease, caused by the kinetoplastid parasite Trypanosoma cruzi, is a human tropical illness mainly present in Latin America. The therapies available against this disease are far from ideal. Proteases pathogenic protozoan have been considered as good drug target candidates. T. cruzi acidic M17 leucyl-aminopeptidase (TcLAP) mediates major parasite's activity and expressed all stages. Here, we report inhibition of TcLAP (IC50 = 66.0 ± 13.5 µM) bestatin-like peptidomimetic KBE009. This...

Trypanosoma cruzi, the causative agent of Chagas disease, has a complex life cycle that involves triatomine insects as vectors and mammals hosts. The differentiation epimastigote forms into metacyclic trypomastigotes within insect vector is crucial for parasite's progression. Factors influencing this process, including temperature, pH, nutritional stress, along with specific metabolite availability, play pivotal role. Amino acids like proline, histidine, glutamine support cell...

Abstract During its complex life cycle, Trypanosoma cruzi colonizes different niches in insect and mammalian hosts. This characteristic determined the types of parasites that adapted to face challenging environmental cues. The primary challenge, particularly stages, is poor nutrient availability. These T. stages could be exposed fatty acids originating from degradation perimicrovillar membrane. In this study, we revisit metabolic fate acid breakdown . Herein, show during parasite...
