Atherosclerosis is associated with alterations in nitric oxide (NO)/cGMP signaling. In early stages of the disease, inflammatory and possibly other cells produce reactive oxygen species that scavenge vasoprotective NO. addition to oxidative stress, expression activity enzymes downstream NO formation may also be affected. Here, we show aortas chronically hypercholesterolemic rabbits (a model late-stage atherosclerosis), both subunits specific receptor soluble guanylyl cyclase (sGC) were...
The mechanism(s) of action responsible for the beneficial effects phosphodiesterase 5 (PDE5) inhibitors including sildenafil on lower urinary tract symptoms suggestive benign prostate hyperplasia are unclear. In particular, role NO-cGMP signalling pathway in regulating human bladder dome smooth muscle relaxation is questionable. Thus, we assessed ability a PDE5 inhibitor, sildenafil, to relax such tissue, and identified pathways involved this relaxation.
Erectile dysfunction is highly prevalent in type II diabetes mellitus. Low intensity extracorporeal shock wave therapy improves erectile function patients with of vasculogenic origin, including diabetes. However, its mode action remains unknown. We investigated the effects low compared to or combined sildenafil on a mellitus model. Our purpose was test our hypothesis targeting cavernous nitric oxide/cyclic guanosine monophosphate pathway.GK rats, validated model mellitus, and age matched...
Hypertensive men have a higher prevalence of erectile dysfunction (ED) than the general population. Experimental evidence ED in hypertensive animals is scarce. This study evaluates function spontaneously rats (SHR) and age-matched normotensive Wistar-Kyoto (WKY) vivo by increase intracavernosal pressure after electrical stimulation cavernous nerve (CN) isometric tension studies on corporal strips. Frequency-dependent responses to CN stimulations were reduced SHR. Phenylephrine induced lower...
Background —We examined the implications of iNOS in atherosclerosis progression using selective inducible NO synthase (iNOS) inhibitor N -iminoethyl- l -lysine (L-NIL) hypercholesterolemic rabbits. Methods and Results —Nine rabbits were fed a 0.3% cholesterol diet for 24 weeks (Baseline group); 25 animals maintained on treated 12 extra with L-NIL (5 mg · kg − 1 d , group, n=8), vehicle (Saline n=9), or -arginine (2.25%, L-Arg n=8). In abdominal aortas Saline rabbits, lesions (53.7±5.7%,...
Erectile dysfunction (ED) is another manifestation of vascular disease. We evaluated the natural history ED in spontaneously hypertensive rat (SHR) and respective participation associated pathophysiological modifications, i.e., endothelial tissue remodeling. SHR their normotensive counterparts [Wistar-Kyoto rats (WKY)] 6, 12, 24 wk age (n = 12) were used to evaluate erectile function, aortic reactivity, responses are reduced at all ages (P < 0.001). In both tissues WKY, relaxations ACh...
To characterize the spontaneous contractile activity (SCA) developed by detrusor from patients with neurogenic overactivity (NDO) because alteration of properties plays a critical role in pathogenesis overactivity, as well to evaluate K(ATP) and K(Ca) channels on this SCA these regulate many species, including humans without overactive bladder (OAB).Human samples were obtained 44 undergoing cystectomy for cancer no known OAB symptoms 38 suffering urodynamically diagnosed NDO. Detrusor strips...
Because insulin resistance is inevitably associated with cardiovascular complications, there a need to further investigate the potential involvement of oxidative stress and cyclo-oxygenase (COX) pathway in vascular modifications this pathological context. Endothelial function was evaluated control fructose-fed rats (FFR) by i) vitro study endothelium-dependent -independent relaxations aortic rings, ii) vivo telemetric evaluation pressor response norepinephrine. After 9 weeks diet, FFR...
What's known on the subject? and What does study add? Lower urinary tract symptoms (LUTS) resulting from benign prostatic hyperplasia (BPH) erectile dysfunction (ED) are common problems in aging male population. Moreover, several recent studies have shown that ED is closely associated with presence severity of LUTS independently co-morbidities. However, pathophysiological mechanisms linking LUTS/BPH remain largely unexplored. The major difficulty studying such relationships between LUTS/BPH,...
