The purpose of this study was to formulate a reservoir-type transdermal delivery system (TDS) for 2,3,5,6-tetramethylpyrazine (TMP) enable the sufficient dose through human skin achieve an effective therapeutic plasma concentration. To improve penetration TMP in TDS, several chemical enhancers were investigated using vitro rat dorsal permeation studies. Eucalyptus oil found enhance greatest extent, with optimal concentration being 5% and flux 542.6 ± 49.7 μg/cm2/h, which 4.5-fold greater...

The intestinal permeability of forskolin was investigated using a single pass perfusion (SPIP) technique in rats. SPIP performed different segments (duodenum, jejunum, ileum, and colon) with three concentrations (11.90, 29.75, 59.90 µg/mL). investigations adsorption stability were to ensure that the disappearance from perfusate due absorption. results study indicated could be absorbed all intestine. effective (P (eff)) range drugs high permeability. P (eff) highest duodenum as compared other...

A major challenge of orally disintegrating tablet (ODT) development is predicting its bioequivalence to corresponding marketed product. Therefore, comparing ODT dissolution profiles those the product very important. The objective this study was develop a 5.2-mg montelukast sodium (MS) with similar profile that chewable tablet. Dissolution were examined in different media screen each formulation. We found MS from ODTs acidic medium heavily depended on manufacturing methods. All prepared using...

Deep eutectic solvents (DESs) are regarded as promising to extract chemicals from plant materials. In this study, a DES-based microwave-assisted extraction (MAE) method was developed for the chromatographic analysis of four bioactive flavonoids in Spirodela polyrrhiza. Chromatographic separation achieved on Promosil C18-column. Prior HPLC analysis, were rapidly extracted by DES-MAE process using choline chloride/levulinic acid (1:2, mol/mol) solvent. The parameters optimized response surface...

A novel reservoir-type transdermal system of 2,3,5,6-tetramethylpyrazine (TMP) was developed containing eucalyptus oil as a penetration enhancer. The single and multiple-dose pharmacokinetic profiles TMP administrated by patch were characterized in healthy volunteers using an vivo, randomized, open-label, two-way crossover design. 2,3,5,6-Tetramethylpyrazine phosphate (TMPP) oral tablets chosen reference. Following single/multiple administration 200/100 mg TMPP tablets, Cmax 1284/613.5 ng/mL...

The pharmacokinetics of bulleyaconitine A (BLA) after a single dose 0.2 mg intramuscular injection was evaluated in healthy volunteers. Physical exam, vital signs, clinical laboratory tests and electrocardiogram measurements were monitored to assess the safety tolerance drug. plasma levels BLA serial samples, collected over 15 h, measured by validated high-performance liquid chromatography (HPLC)-electrospray ionization tandem mass spectrometry (MS-MS) method. It demonstrated that absorbed...

The degradation kinetics of forskolin in aqueous solution was investigated qualitatively and quantitatively. Two products were isolated identified as isoforskolin D by liquid chromatography–tandem mass spectrometry (LC–MS/MS) nuclear magnetic resonance (NMR) spectroscopy. A stability-indicating high-performance chromatography (HPLC) method developed validated for the quantification its products. Chromatographic separation performed on a Luna C18 column with acetonitrile–water (65:35, v/v)...

Abstract An HPLC method was established for preparation of scutellarein from scutellarin using an enzymolysis reaction. The aglycone identified as with its chromatogram, UV, LC‐MS, and 1H‐NMR spectra. Acid hydrolysis tried in preliminary experiments, but adopted after optimization reaction conditions. High purity (content 98.04%∼98.36%) prepared this way the first time, which could be used a working reference substance.

Hexyl aminolevulinate (HAL) is a lipophilic derivative of 5-aminolevulinic acid (5-ALA) and can induce more protoporphyrin IX (PpIX) formation stronger fluorescence intensity (FI) than 5-ALA, which will greatly facilitate photodynamic diagnosis therapy. The main drawback HAL its low solubility in neutral aqueous media. In this study, surfactants were used to increase the cell culture medium serum, followed by vitro measurement human pancreatic cancer cells (SW1990) vivo detection...
