Abstract Pulmonary hypertension (PH) is a syndrome complex that accompanies number of diseases different etiologies, associated with basic mechanisms structural and functional changes the pulmonary circulation vessels revealed pressure increasing in artery. The are main limiting factor determining prognosis patients PH. Thickening irreversible deposition collagen artery branches walls leads to rapid disease progression therapy effectiveness decreasing. In this regard, histological...

Aim . To develop a model of chronic thromboembolic pulmonary hypertension (CTEPH) in rats by embolization the vascular bed with microencapsulated fibrin (MF). Materials and methods Microencapsulated (MF) was prepared encapsulating particles smaller than 71 μm sodium alginate. Non-encapsulated particle size 71–200 µm used as an alternative embolic particle. Modeling performed on male Wistar rats. The animals were divided into 4 groups. Intact (INT) ( n = 7) administered normal saline...

Summary This study aimed to investigate the effect of bone marrow‐ and adipose tissue‐derived mesenchymal stem cell ( BM ‐ MSC AD respectively) transplantation on left ventricular function infarct area IA ) in rat model ischaemic heart failure. In anaesthetized Wistar rats, coronary artery LCA was occluded for 40 min with subsequent reperfusion 7 days. Seven days following surgery, animals occlusion/reperfusion were randomized into three groups: (i) Controls received intramyocardial...

Summary A major translational barrier to the use of stem cell ( SC )‐based therapy in patients with myocardial infarction MI ) is lack a clear understanding mechanism(s) underlying cardioprotective effect s. Numerous paracrine factors from s may account for reduction infarct size, but salvage associated transdifferentiation into vascular cells as well cardiomyocyte‐like be involved too. In this study, bone marrow‐derived rat mesenchymal MSC s) were microencapsulated alginate preventing...

Chronic thromboembolic pulmonary hypertension (CTEPH) is a rare and life-threatening complication of embolism. As existing animal models CTEPH do not fully recapitulate complex disease pathophysiology, we report new rat model for evoked by repetitive embolization the distal artery branches with partially biodegradable alginate microspheres (MSs). MSs (180 ± 28 μm) were intravenously administered eight times at 4-day intervals; control animals received saline. The validity was confirmed using...

According to global health statistics, respiratory diseases, together with infectious complications and hereditary lung rank as the third leading cause of death. Today, transplantation (LTx) is a well-recognized modality treatment for end-stage chronic disease. However, number LTx surgeries performed much lower than other solid organs. This due high requirements potential donor characteristics graft, reflecting efficiency gas exchange function. Non-compliance selection criteria leads...

Chronic thromboembolic pulmonary hypertension (CTEPH) is a rare complication of acute embolism with poor clinical outcomes. Therapeutic approaches to prevention fibrotic remodeling the vascular bed in CTEPH are limited. In this work, we tested hypothesis that Janus kinase 1/2 (JAK1/2) inhibition ruxolitinib might prevent and attenuate rat model. was induced by repeated embolization artery partially biodegradable 180 ± 30 μm alginate microspheres. An entire cycle of preclinical studies 4-[(3-ethoxy-3-oxopropanoyl)amino]benzoic acid (etmaben) exhibiting cardiotropic action was carried out. Its potential mechanism and pharmacological effects, general toxic effects in acute chronic experiments, specific types toxicity (allergenicity, immunotoxicity, reproductive toxicity, mutagenicity), pharmacokinetics were studied. The drug found to have a pronounced effect positive safety profile, which justifies the launch phase I clinical trials.
