Vaccine efforts to combat the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), which is responsible for current disease 2019 (COVID-19) pandemic, are focused on SARS-CoV-2 spike glycoprotein, primary target neutralizing antibodies. We performed cryo-election microscopy and site-specific glycan analysis of one leading subunit vaccine candidates from Novavax, based a full-length protein formulated in polysorbate 80 detergent. Our studies reveal stable prefusion conformation...

Enveloped viruses hijack not only the host translation processes, but also its glycosylation machinery, and to a variable extent cover viral surface proteins with tolerogenic host-like structures. SARS-CoV-2 protein S presents as trimer on is covered by dense shield of N-linked glycans, few O-glycosites have been reported. The location O-glycans controlled large family initiating enzymes expression in cells tissues hence difficult predict. Here, we used our well-established O-glycoproteomic...

The dense array of N-linked glycans on the HIV-1 envelope glycoprotein (Env), known as "glycan shield," is a key determinant immunogenicity, yet intrinsic heterogeneity confounds typical structure-function analysis. Here, we present an integrated approach single-particle electron cryomicroscopy (cryo-EM), computational modeling, and site-specific mass spectrometry (MS) to probe glycan shield structure behavior at multiple levels. We found that dynamics lead extensive network interglycan...

The global incidence of metabolic dysfunction-associated fatty liver disease (MAFLD) is increasing annually, which has become a major public-health concern. MAFLD typically associated with obesity, hyperlipemia, or syndrome. Dietary induction one the most common methods for preparing animal models MAFLD. Viruses can evade the host immune system by displaying numerous glycans on their surface "spike-proteins" that cover epitopes. We have developed an ultrasensitive "single-pot" method to assess glycan occupancy and extent of processing from high-mannose complex forms at each N-glycosylation site. Though aimed characterizing glycosylation viral spike-proteins as potential vaccines, this is applicable for analysis site-specific any glycoprotein.
