We investigated the effects of bicarbonate on growth several different bacteria as well its biofilm formation and intracellular cAMP concentration in Pseudomonas aeruginosa. Biofilm was examined 96-well plates, with or without bicarbonate. The production measured by a commercial assay kit. found that NaHCO3 (100 mmol l-1) significantly inhibited, whereas NaCl did not influence planktonic bacteria. MIC MBC measurements indicated effect HCO3- is bacteriostatic rather than bactericidal....

Background: 3D printing is a rapidly developing technology in the healthcare industry and dentistry. Its application clearly shows that this area of digital dentistry has potential for everyday usage across all fields, including prosthodontics, orthodontics, maxillofacial surgery, oral implantology. However, despite gaining ground, there lack information about how specialists (dentists dental technicians) use additive technology. Our research group aimed to investigate impact social media on...

Primary biliary cirrhosis (PBC) is a disorder of unknown origin with autoimmune features. Recently, impaired secretion bicarbonate has been shown in patients PBC. Here we have investigated whether bile duct epithelial cells isolated from PBC exhibit defects transepithelial transport by analyzing the activities 2 ion exchangers, Cl − /HCO 3 anion exchanger (AE2) and Na + /H (NHE) cholangiocytes. AE2 NHE were studied basal conditions after stimulation cyclic adenosine monophosphate (cAMP)...

Background/Aims: ATP-gated P2X4 purinergic receptors (P2X4Rs) are cation channels with important roles in diverse cell types. To date, lack of specific inhibitors has hampered investigations on P2X4Rs. Recently, the benzodiazepine derivative, 5-BDBD been proposed to selectively inhibit However, limited evidences currently available its inhibitory properties. Thus, we aimed characterize effects recombinant human Methods: We investigated ATP-induced intracellular Ca2+ signals and whole ion...

Purinergic receptor stimulation has potential therapeutic effects for cystic fibrosis (CF). Thus, we explored roles P2Y and P2X receptors in stably increasing [Ca2+]i human CF (IB3-1) non-CF (16HBE14o−) airway epithelial cells. Cytosolic Ca2+ was measured by fluorospectrometry using the fluorescent dye Fura-2/AM. Expression of (P2XR) subtypes assessed immunoblotting biotinylation. In IB3-1 cells, ATP other agonists caused only a transient increase derived from intracellular stores Na+-rich...

Cystic fibrosis (CF) is caused by defective cyclic AMP-dependent cystic transmembrane conductance regulator Cl- channels. Thus, CF epithelia fail to transport and water. A postulated therapeutic avenue in activation of alternative Ca2+-dependent We hypothesized that stimulation Ca2+ entry from the extracellular space could trigger a sustained signal activate Cytosolic [Ca2+]i was measured non-polarized human (IB3-1) non-CF (16HBE14o-) airway epithelial cells. Biliary epithelial cells (cholangiocytes) modulate bile fluidity and alkalinity absorbing and/or secreting fluid electrolytes, particularly HCO 3 − Cl . Mechanisms responsible for transepithelial H + /HCO secretion in human cholangiocytes are largely unknown. Biliary epithelial cells (cholangiocytes) are responsible for rapid regulation of bile volume and alkalinity. Secretin other hormones raising intracellular cyclic adenosine monophosphate (cAMP) concentrations promote biliary HCO3 secretion by stimulating apical Cl- channels Cl-/HCO3- exchange (AE2). Cholangiocyte ion transport may also be stimulated locally acting mediators; example, 5'-triphosphate (ATP), a secretagogue that can released into the hepatocytes cholangiocytes, activates...
