We describe a system to evaluate the function of lymphocyte-specific and generally expressed genes in differentiation and/or lymphocytes. RAG-2 (recombination-activating gene 2)-deficient mice have no mature B T lymphocytes due inability initiate VDJ recombination. Blastocysts from RAG-2-deficient generate animals with cells following implantation into foster mothers. However, injection normal ES blastocysts leads generation somatic chimeras all which derive injected (referred as blastocyst...

Leptin, an adipocytokine that suppresses appetite and may regulate neuroendocrine pathways, is low in undernourished states like anorexia nervosa (AN). Although leptin exhibits pulsatility, secretory characteristics have not been well described adolescents AN, the contribution of hypoleptinemia to increased growth hormone (GH) cortisol AN has explored. We hypothesized reflects decreased basal pulsatile secretion predict GH levels. Sampling for leptin, GH, cortisol, ghrelin was performed...

Ghrelin is an orexigenic peptide and a growth hormone (GH) secretagogue. Secretory dynamics of ghrelin have not been characterized in adolescents with anorexia nervosa (AN). We hypothesized that, compared healthy adolescents, girls AN would increased concentrations measured over 12 h nocturnal sampling from basal pulsatile secretion, endogenous independently predict GH cortisol. examined concentration secretory 22 18 12-18 yr old. Associations between ghrelin, various hormones, measures...

Anorexia nervosa (AN) in adolescents is associated with low bone mineral density (BMD) and increases ghrelin secretion, an orexigenic GH secretagogue that stimulates osteoblast proliferation vitro. We hypothesized may have independent effects on AN adolescents. STUDY DESIGN, SUBJECTS, AND OUTCOME MEASURES: Frequent sampling was performed overnight every 30 min for 12 h 23 adolescent girls aged 12-18 yr 21 controls of comparable maturity. Ghrelin, leptin, cortisol, secretion were examined...

We have isolated cDNA clones complementary to a truncated immunoglobulin heavy-chain C epsilon RNA transcript previously found be induced in B lymphoid cells by treatment with lipopolysaccharide (LPS) combined interleukin-4 (IL-4). demonstrate that this initiates from promoter upstream of the germ line class-switch recombination region (S region). The major contains small 5' exon contributed sequences S spliced normal exons. Treatment splenic LPS plus IL-4 induces expression transcripts...

The objective of this article is to address the challenges physical sample retrieval from all locations along patients' gastrointestinal (GI) tracts for studying interactions between GI microbial communities (microbiomes) and host immune systems. We propose a novel tetherless, blindly controlled, magnetically actuated capsule noninvasive sampling microbiomes liquid digesta. mesoscale capsule, soft mobile robot platform, made two permanent magnets encapsulated in elastomer remotely by an...

Objective To assess site of disease on preoperative computed tomography ( CT ) to predict surgical debulking in patients with ovarian cancer. Design Two‐phase retrospective cohort study. Setting West London Gynaecological Cancer Centre, UK . Population Women stage 3 or 4, ovarian, fallopian primary peritoneal cancer undergoing cytoreductive surgery. Methods Preoperative images were reviewed by experienced radiologists the presence absence at predetermined sites. Multivariable stepwise...

The authors report on three patients who received intra-articular corticosteroid injections (IACI) in the small joints of their hands for symptomatic relief local inflammation. In these cases radiographs showed that among complications IACI are periarticular soft-tissue calcifications. Evidence suggests calcifications composed hydroxyapatite crystals, which may themselves produce inflammatory arthritis.

Abstract High-throughput sequencing-based methods for bulked segregant analysis (BSA) allow the rapid identification of genetic markers associated with traits interest. BSA studies have successfully identified qualitative (binary) and quantitative trait loci (QTLs) using QTL mapping. However, most require population structures that fit models available a reference genome. Instead, high-throughput short-read sequencing can be combined k-mers (BSA-k-mer) to map appear refractory standard...
