During the SARS-CoV-2 pandemic, novel and traditional vaccine strategies have been deployed globally. We investigated whether antibodies stimulated by mRNA vaccination (BNT162b2), including third-dose boosting, differ from those generated infection or adenoviral (ChAdOx1-S Gam-COVID-Vac) inactivated viral (BBIBP-CorV) vaccines. analyzed human lymph nodes after for correlates of serological differences. Antibody breadth against variants is lower compared with all vaccines evaluated but...
Duchenne muscular dystrophy (DMD) causes profound and progressive muscle weakness loss, resulting in early death. DMD is usually caused by frameshifting deletions the gene DMD, which leads to absence of dystrophin protein. Dystrophin binds F-actin components dystrophin-associated glycoprotein complex protects sarcolemma from contraction-induced injury. Antisense oligonucleotide-mediated exon skipping a promising therapeutic approach aimed at restoring reading frame allowing expression an...
TCR engagement triggers the polarized recruitment of membrane, actin, and transducer assemblies within T cell-APC contact that amplify specify signaling cascades effector activity. We report caveolin-1, a scaffold regulates polarity in nonlymphoid cells, is required for optimal TCR-induced actin polymerization, synaptic membrane raft polarity, function CD8, but not CD4, cells. In CD8(+) caveolin-1 ablation selectively impaired NFAT-dependent NFATc1 cytokine gene expression, whereas...
Abstract Despite recent advances in therapeutic approaches, patients with MLL-rearranged leukemia still have poor outcomes. Here, we find that the RNA-binding protein IGF2BP3, which is overexpressed MLL-translocated leukemia, strongly amplifies MLL-Af4-mediated leukemogenesis. Deletion of Igf2bp3 significantly increases survival mice MLL-Af4-driven and greatly attenuates disease, a minimal impact on baseline hematopoiesis. At cellular level, MLL-Af4 leukemia-initiating cells require for...
Epstein-Barr virus (EBV) is a potent carcinogen linked to hematologic and solid malignancies causes significant global morbidity mortality. Therapy using allogeneic EBV-specific lymphocytes shows promise in certain populations, but the impact of EBV genome variation on these strategies remains unexplored. To address this, we sequenced 217 genomes, including from Guatemala, Peru, Malawi, Taiwan, analyzed them alongside 1307 publicly available genomes cancer, nonmalignant diseases, healthy...
Despite advances in treatments, multiple myeloma (MM) remains an incurable cancer where relapse is common. We developed a circulating tumor DNA (ctDNA) approach order to characterize genomics, monitor treatment response, and detect early MM. By sequencing 412 specimens from 64 patients with newly diagnosed or relapsed/refractory disease, we demonstrate the correlation between ctDNA key clinical biomarkers, as well patient outcomes. further extend our simultaneously track CAR-specific...
Background The polarized reorganization of the T cell membrane and intracellular signaling molecules in response to receptor (TCR) engagement has been implicated modulation development effector responses. In siRNA-based studies Dlg1, a MAGUK scaffold protein member Scribble polarity complex, shown play role TCR signal specificity, however Dlg1 function vivo remains unclear. Methodology/Principal Findings Here we present combined data from three independently-derived dlg1-knockout mouse...
Abstract Inadequate diagnostics compromise cancer care across lower- and middle-income countries (LMICs). We hypothesized that an inexpensive gene expression assay using paraffin-embedded biopsy specimens from LMICs could distinguish lymphoma subtypes without pathologist input. reviewed all obtained at the Instituto de Cancerología y Hospital Dr. Bernardo Del Valle in Guatemala City between 2006 2018 for suspicion of lymphoma. Diagnoses were established based on World Health Organization...
Abstract Lymphomas vary in terms of clinical behavior, morphology, and response to therapies thus accurate classification is essential for appropriate management patients. In this study, using a set 670 cases lymphoma obtained from center Guatemala City, we propose an interpretable machine learning method, LymphoML, subtyping into eight diagnostic categories. LymphoML sequentially applies steps (1) object segmentation extract nuclei, cells, cytoplasm hematoxylin eosin (H&E)-stained...
Functionally diverse CD8+ T cells develop in response to antigenic stimulation with differing capacities couple TCR engagement downstream signals and functions. However, mechanisms of diversifying signaling are largely uncharacterized. Here we identified two alternative splice variants scaffold protein Dlg1, Dlg1AB Dlg1B, that diversify regulate p38 –dependent –independent effector functions cells. Dlg1AB, but not Dlg1B associated Lck, coupling activation proinflammatory cytokine production....
This study describes the psychometric properties of a Spanish version Shortened Outcome Questionnaire (OQ-30.2, Lambert et al., 2004), made for Chilean population. The questionnaire was administered to 546 patients in an outpatient mental health clinic and 100 non-clinical adults. Results indicated that measure has good internal consistency (α=0.90) sensitivity change. Furthermore, results were similar those obtained its original version, making possible use context.
Measurable residual disease flow cytometry (MRD-FC) and molecular studies are the most sensitive methods for detecting malignant populations after therapy TP53-mutated acute myeloid leukemia myelodysplastic neoplasms (TP53+ AML/MDS). However, their sensitivity is limited in suboptimal aspirates or when immunophenotype of neoplastic blasts overlaps with erythroids normal maturing cells. In this study, we set out to determine if p53 immunohistochemistry (IHC) correlates MRD-FC next-generation...
CD8(+) T cells respond to TCR stimulation by producing proinflammatory cytokines, and destroying infected or malignant through the production release of cytotoxic granules. Scaffold protein Discs large homolog 1 (Dlg1) specifies TCR-dependent functions channeling proximal signals toward activation p38-dependent cytokine gene expression and/or p38-independent granule release. Two Dlg1 variants are expressed in via alternative splicing, Dlg1AB Dlg1B, which have differing abilities coordinate...

Mature plasmacytoid dendritic cell proliferations (MPDCPs) are clonal, non-malignant pDC that have been reported to occur in association with myeloid neoplasms such as CMML, AML (pDC-AML), and, rarely, MDS or MPNs. Here we report the first case of a MPDCP associated T-lymphoblastic leukemia (T-ALL), lymphoid neoplasm. The this involved ~50% bone marrow, was found nodular aggregates, expressed CD123, CD4, and CD303, lacked CD56 TCL1 expression. In addition, CD34 TdT but showed aberrant...
Castleman disease (CD) is a rare lymphoproliferative disorder with distinct clinical subtypes. However, our understanding of the underlying pathogenesis particular subtypes CD remains unclear. While characteristic morphologic changes within UCD, including occasional cases overgrowth spindled stromal and follicular dendritic cells have been described, nature origin these spindle remain elusive. Few reports suggested that in UCD are clonally neoplastic may be fibroblastic reticular cell (FRC)...
