Monacolin K is a secondary metabolite synthesized by polyketide synthases (PKS) from Monascus, and it has the same structure as lovastatin, which mainly produced Aspergillus terreus. In present study, bacterial artificial chromosome (BAC) clone, mps01, was screened BAC library constructed Monascus pilosus BCRC38072 genomic DNA. The putative monacolin biosynthetic gene cluster found within 42 kb region in mps01 clone. deduced amino acid sequences encoded nine genes designated mokA−mokI, share...

In the present study, IL-1β cDNA was identified and analyzed from largemouth bass (Micropterus salmoides). Full length mRNA obtained using Rapid Amplification of Ends (RACE), which contains 78 bp 3′-UTR, a 455 5′-UTR, an open reading frame (ORF) 702 coding for 233 amino acid residues. The molecular weight theoretical isoelectric point protein predicted to be 26.7 kDa 6.08 respectively. A phylogenetic analysis showed close relation IL-1βs striped trumpeter (Latris lineata), Chinese perch...

Citrinin, a hepato-nephrotoxic compound to humans, can be produced by the food fermentation microorganisms Monascus spp. In this study, we investigated distribution of mycotoxin citrinin biosynthesis genes in 18 strains. The results show that acyl-transferase and keto-synthase domains pksCT gene encoding polyketide synthase were found purpureus, kaoliang, sanguineus. Furthermore, ctnA gene, major activator for biosynthesis, was M. purpureus but absent orf3 oxygenase, located between ctnA,...

The filamentous fungus Monascus pilosus was genetically transformed with a reporter plasmid, pMS-1.5hp, by aurintricarboxylic acid (ATA) treatment to obtain an efficient red-pigment producing mutant. transformation efficiency of higher the ATA-treatment than either non-restriction-enzyme-mediated integration (REMI) or REMI method. This valid and convenient random mutagenesis method shows that ATA can be applied in fungi for genetic transformation.
