ABSTRACT Objectives Epithelial cells are an important part of the pathomechanism in chronic rhinosinusitis with nasal polyps. It is therefore essential to establish a robust method for isolation and culture epithelial from polyps enable further research. In this study, feasibility outgrowth technique was evaluated. Methods The performed isolate cells. Proliferation evaluated up 3rd passage. were identified differentiation proliferation using flow cytometry anti-cytokeratin, anti-p63,...
Epithelial cells may play an important role in the pathologic process of chronic rhinosinusitis with nasal polyps. Therefore, providing epithelial from a biobank could greatly contribute to further research. In present work, isolation long-term cryopreserved tissue is demonstrated. Polyp tissues were commercially available freezing medium dimethyl sulfoxide and stored liquid nitrogen. The outgrowth proliferation evaluated compared that fresh tissue. Flow cytometric analysis anti-cytokeratin,...
Abstract Background Epithelial cells are an important part of the pathomechanism in chronic rhinosinusitis with nasal polyps. It is therefore essential to establish a robust method for isolation and culture epithelial from polyps enable further research. In this study, feasibility outgrowth technique was evaluated. Results Using technique, could be isolated all tissue samples. Isolated showed proliferation rate approximately 7- 23-fold every 6 days up 3rd passage. Over 97% were shown...
Background Epithelial cells (EC) are an important component of the pathomechanism type 2 inflammation in chronic rhinosinusitis with nasal polyps. Therefore, it is to establish a robust method for isolation and culture EC from polyps enable further investigation. In this study, we evaluated feasibility outgrowth technique isolate
Background Chronic rhinosinusitis with nasal polyps is commonly caused by a complex Th-2 cell-based inflammatory response in Western countries. However, the exact pathomechanism still largely unknown. In order to enable further sufficient investigations and new insights, it necessary develop an investigation tool that can reflect this complexity. Research question present study, potential of organ culture tissue from as was evaluated.
The biological drug dupilumab, an anti-interleukin-4α receptor antibody, approved in 2019, can be assumed to highly effective patients with chronic rhinosinusitis nasal polyps (CRSwNP). However, the mechanism of action downstream blockade is largely unknown.
Chronic rhinosinusitis with nasal polyps (CRSwNP) is a chronic inflammatory disease of the upper respiratory tract. The epithelial cells are an important part pathomechanism, so that they may play essential role for secretion pro-inflammatory cytokines and remodeling mucosa.
