It has been postulated that during human fetal development, all cells of the lung epithelium derive from embryonic, endodermal, NK2 homeobox 1-expressing (NKX2-1+) precursor cells. However, this hypothesis not formally tested owing to an inability purify or track these progenitors for detailed characterization. Here we have engineered and developmentally differentiated NKX2-1GFP reporter pluripotent stem (PSCs) in vitro generate isolate primordial express NKX2-1 but are initially devoid...

Abstract Efficient generation of human induced pluripotent stem cell (hiPSC)-derived intestinal organoids (HIOs) would facilitate the development in vitro models for a variety diseases that affect gastrointestinal tract, such as inflammatory bowel disease or Cystic Fibrosis. Here, we report directed differentiation protocol mesenchyme-free HIOs can be primed towards more colonic proximal lineages serum-free defined conditions. Using CDX2 eGFP iPSC knock-in reporter line to track emergence...

The in vitro directed differentiation of pluripotent stem cells (PSCs) through stimulation developmental signaling pathways can generate mature somatic cell types for basic laboratory studies or regenerative therapies. However, there has been significant uncertainty regarding how to separately derive lung vs. thyroid epithelial lineages, since these two each originate from Nkx2-1+ foregut progenitors, and the minimal claimed regulate their distinct lineage specification vivo have varied...

Lung epithelial lineages have been difficult to maintain in pure form vitro, and lineage-specific reporters proven invaluable for monitoring their emergence from cultured pluripotent stem cells (PSCs). However, reporter constructs tracking proximal airway generated PSCs not previously available, limiting the characterization of these cells. Here, we engineer mouse human PSC lines carrying secretory lineage that facilitate tracking, purification, profiling this lung subtype. Through bulk...

Regenerative medicine techniques are being widely investigated for organs without regenerative capacity, such as lung. Whether pluripotent stem cell derived thyroid organoids will be used in the future a approach to treat hypothyroidism is unclear. In context of an easily available, very cheap and non-invasive substitutive therapy with levothyroxine, cost-risk balance clearly speaks against novel approach. However, pneumocyte follicular precursors share key transcription factor, NKX2-1,...

Melioidosis, an emerging infectious disease in India is well known for its protean clinical manifestations. Definitive diagnosis can be made by isolation of non-fermentative Gram-negative bacilli from specimens and identification the isolate at species level. We report a case disseminated melioidosis with lymphadenitis prostatic abscess. Burkholderia pseudomallei was isolated bone marrow, pus aspirate, blood urine, patient responded to treatment.
