The mechanism by which ZIKV causes a range of neurological complications, especially congenital microcephaly, is not well understood. fact that microcephaly associated with Asian lineage strains raises the question why this was discovered earlier. One possible explanation and African differ in their abilities to infect cells CNS cause neurodevelopmental problems. Here, we show induce cell death human neural progenitor cells—which are important target development microcephaly—less efficiently...
The recent epidemic of Zika virus (ZIKV) in the Americas and its association with fetal neurological complications has shown need to develop a treatment. Repurposing drugs that are already FDA approved or clinical development may shorten drug timelines case emerging viral diseases like ZIKV. Initial studies have conflicting results when testing sofosbuvir developed for treatment infections another Flaviviridae virus, hepatitis C virus. We hypothesized could be explained by differences...
Zika virus (ZIKV) infection is typically characterized by a mild disease presenting with fever, maculopapular rash, headache, fatigue, myalgia and arthralgia. A recent animal study found that ZIKV-infected pregnant Ifnar-/-mice developed vascular damage in the placenta reduced amount of fetal capillaries. Moreover, ZIKV causes segmental thrombosis umbilical cord rhesus macaques. Furthermore, several case reports suggest cause coagulation disorders. These results could an alteration host...
In 2015-2016, a Zika virus (ZIKV) outbreak occurred in the Americas. 2017, we conducted ZIKV serosurvey Suriname which 770 participants were recruited from 1 urban area and 2 rural villages tropical rainforest. All collected samples tested for presence of antibodies using immunoglobulin G enzyme-linked immunosorbent assay neutralization assay. We found that 35.1% had neutralizing against ZIKV. remote village rainforest, 24.5% ZIKV, suggesting was widely spread across Suriname.
Abstract Zika virus (ZIKV) infection is typically characterized by a mild self-limiting disease presenting with fever, rash, myalgia and arthralgia severe fetal complications during pregnancy such as microcephaly, subcortical calcifications arthrogyropsis. Virus-induced due to perturbed osteoblast function has been described for other arboviruses. In case of ZIKV infection, the role osteoblasts in pathogenesis bone related pathology remains unknown. Here, we study effect on differentiation,...
Bone-related complications are commonly reported following arbovirus infection. These arboviruses known to disturb bone-remodeling and induce inflammatory bone loss via increased activity of resorbing osteoclasts (OCs). We previously showed that Zika virus (ZIKV) could the function forming osteoblasts, but susceptibility OCs ZIKV infection is not known. Here, we investigated effect on osteoclastogenesis report pre- early with significantly reduced osteoclast formation resorption....
During the 2015-2016 outbreak of Zika virus (ZIKV) in Americas, a previously unknown severe complication ZIKV infection during pregnancy resulting birth defects was reported. Since occurred regions that were highly endemic for related dengue (DENV), it speculated antibody-dependent enhancement (ADE) infection, caused by presence cross-reactive DENV antibodies, could contribute to disease severity. Emerging evidence indicates that, while vitro models can show ADE does not seem congenital...
Abstract Arboviruses target bone forming osteoblasts and perturb remodeling via paracrine factors. We previously reported that Zika virus (ZIKV) infection of early‐stage human mesenchymal stromal cells (MSCs) inhibited the osteogenic lineage commitment MSCs. To understand physiological interplay between development ZIKV pathogenesis, we employed a primary in vitro model to examine biological responses MSCs at different stages osteogenesis. Precommitted were infected late stage stimulation...
