Pitx2, a bicoid-related homeobox gene, is involved in Rieger's syndrome and the left-right (L-R) asymmetrical pattern formation body plan. In order to define genomic structure roles of we analyzed generated Pitx2-deficient mice with lacZ gene homeobox-containing exon Pitx2. We were able show that among three isoforms Pitx2c shows expression whereas Pitx2a, Pitx2b symmetrical expression. Pitx2(-)(/)(-) embryos there was an increase mesodermal cells distal end left lateral wall amnion...
Leptin acts as a potent inhibitory factor against obesity by regulating energy expenditure, food intake, and adiposity. The obese diabetic db / mouse, which has defects in leptin receptor, displays enhanced neural responses elevated behavioral preference to sweet stimuli. Here, we show the effects of on peripheral taste system. An administration into lean mice suppressed nerves (chorda tympani glossopharyngeal) substances (sucrose saccharin) without affecting sour, salty, bitter substances....
Leptin is a hormone that regulates body weight homeostasis mainly via the hypothalamic functional leptin receptor Ob-Rb. Recently, we proposed taste organ new peripheral target for leptin. selectively inhibits mouse cell responses to sweet substances and thereby may act as modulator. The present study further investigated action on system by examining expression of Ob-Rb in cells behavioral leptin-deficient ob/ob, Ob-Rb-deficient db/db mice their normal litter mates. RT-PCR analysis showed...
Taste buds contain ∼60 elongate cells and several basal cells. Elongate comprise three functional taste cell types: I, glial cells; II, bitter/sweet/umami receptor III, sour detectors. Although are continuously renewed, lineage relationships among types ill-defined. Basal have been proposed as bud stem cells, a subset of which express Sonic hedgehog (Shh). However, Shh+ turn over rapidly suggesting that post-mitotic precursors some or all types. To fate map Shh-expressing mice carrying...
Seven lines of genetically modified (GM) maize have been authorized in Japan as foods and feeds imported from the USA. We improved a multiplex PCR method described previous report order to distinguish five GM maize. Genomic DNA was extracted with silica spin column kit, which could reduce experimental time improve safety laboratory potentially environment. sequenced recombinant (r-DNA) introduced into maize, re-designed new primer pairs increase specificity by PCR. A pair for intrinsic zein...
Polymerase Chain Reaction (PCR) techniques are increasingly used for the detection of genetically modified (GM) crops in foods. In this paper, recombinant DNAs introduced into seven lines GM maize, such as Event 176, Bt11, T25, MON810, GA21, DLL25, and MON802, sequenced. On basis obtained sequence, 14 primer pairs segments, promoter, terminator regions, construct genes, were designed. To confirm specificities designed pairs, PCR was performed on genomic extracted from non-GM soy, other...
A method using polymerase chain reaction (PCR) was designed for the detection of genetically modified maize (GM-maize). There are four lines GM-maize imported from United States, and presence recombinant deoxyribonucleic acid (DNA) in could be detected with pairs specific oligonucleotide primers sequences newly introduced genes. The zein gene also as an internal control. This allows each Bt11, Event176, MON810 LIBERTY by to amplify a segment including part exogenously sequence intrinsic...
Leptin is a hormone that regulates food intake, energy expenditure and body weight. Our previous studies have demonstrated the taste organ new peripheral target for leptin in mice. selectively inhibits responses of nerves receptor cells to sweet substances without affecting sour, salty, bitter substances. Still, there no convincing evidence existence receptors (Ob-Rs) cells, especially functional isoform Ob-Rb. We investigated expression 5 different Ob-R isoforms (a-e) 6 STAT (signal...
Neural cell adhesion molecule (NCAM) is a type III marker in the taste buds. In order to clarify of Mash1-expressing cells buds, expression NCAM was examined adult mice comparison with gustducin- and T1r3-expressing cells, using combination immunohistochemistry situ hybridization. About 98% were immunopositive (IP), suggesting that should be categorized as cells. Unexpectedly, small subsets also found NCAM-IP, contradicting previous immunohistochemical studies rats, which gustducin-IP...
Abstract Although embryonic expression of Shh in the fungiform papilla placodes has a critical role patterning, it remains unclear whether its appearance indicates differentiation basal cells taste buds. To examine development cells, , Prox1 and Mash1 was determined anterior tongue soft palate mouse embryos by situ hybridization. In tongue, coexpressed with from beginning at E12.5. expressed band‐like pattern anteriormost region punctate posterior E14.5. The number (21.4 ± 4.3, E14.5)...
A method using polymerase chain reaction (PCR) was designed for the detection of food or ingredients derived from genetically modified soybeans (GMS), imported United States, in a mixture with conventional non-genetically (non-GMS). The presence recombinant deoxyribonucleic acid (DNA) could be detected three different pairs specific oligonucleotide primers sequences introduced genes. soybean intrinsic lectin Le1 gene used as an internal control. results PCR amplification indicated that...
Taste bud cells have a limited lifespan and are continuously replaced just like other epithelial cells. Although there is some evidence that taste buds may arise from the local epithelium, receptor neuronal properties. This implies must be critical stage at which precursor for start to exhibit neural properties during differentiation of The expression neural-specific transcription factors Mash-1 Prox-1 in nervous system transient precedes differentiation. Therefore, we examined epithelium...
Highly homologous DNA elements were found to be shared by the upstream regions of mouse tyrosinase and related protein (TRP‐1) genes. Several nuclear proteins shown bind both these regions. Shared also in 5’ flanking sequences Japanese quail snapping turtle nucleotide scattered like an archipelago human Comparisons between genes gave similar results. On contrary, mammalian (mouse human) nonmammalian (quail turtle) did not show significant homology their In contrast, coding first exons...
