The performance of the LHCb Muon system and its stability across full 2010 data taking with LHC running at ps = 7 TeV energy is studied. optimization detector setting time calibration performed first collisions delivered by described. Particle rates, measured for wide range luminosities beam operation conditions experienced during run, are compared values expected from simulation. space alignment detectors, chamber efficiency, resolution cluster size evaluated. found to be as specifications...
Metformin treatment improved ADPKD disease severity in a relevant, slowly progressive mouse model that recapitulates PKD-associated PKD1 mutation. Relative to controls, metformin reduced kidney weight/body weight, cystic index and BUN levels, while improving GFR, blood pressure anemia. also key inflammatory injury markers, along with cell proliferation markers. These findings suggest several beneficial effects of this model, which may help inform new therapies patients.
Phenotypic heterogeneity has been observed among mesenchymal stem/stromal cell (MSC) populations, but specific genes associated with this variability have not defined. To study question, we analyzed two distinct isogenic MSC populations isolated from umbilical cord blood (UCB1 and UCB2). The use of eliminated differences contributed by genetic background. We characterized these UCB MSCs for morphology, growth kinetics, immunophenotype, potential differentiation. UCB1 displayed faster higher...
Tissue-specific gene expression defines cellular identity and function, but knowledge of early human development is limited, hampering application cell-based therapies. Here we profiled 5 distinct cell types at a single fetal stage, as well chondrocytes 4 stages in vivo 2 during vitro differentiation. Network analysis delineated five tissue-specific modules; these modules chromatin state defined broad similarities cartilage specification maturation vivo, including progressive silencing...
Working with cultured dermal fibroblasts derived from control individuals and patients systemic sclerosis (SSc), we have examined the effects of protein kinase C-δ (PKC-δ) on type I collagen biosynthesis steady-state levels COL1A1 COL3A1 mRNAs. Rottlerin, a specific inhibitor PKC-δ, exerted powerful, dose-dependent inhibition III gene expression in normal SSc cells. Optimal rottlerin concentrations caused 70–90% production, >80% reduction mRNA, >70% mRNA both cell types. In vitro nuclear...
The nuclear emulsion target of the CHORUS detector was exposed to wide-band neutrino beam CERN SPS 27 GeV average energy from 1994 1997. In total, about 100 000 charged-current (CC) interactions with at least one identified muon were located in and fully reconstructed, using newly developed automated scanning systems. Charmed particles searched for by a program recognizing particle decays. observation decay makes it possible select sample very low background minimal kinematical bias. all,...
Skeletal dysplasias (SDs) are caused by abnormal chondrogenesis during cartilage growth plate differentiation. To study early stages of aberrant formation in vitro, we generated the first induced pluripotent stem cells (iPSCs) from fibroblasts an SD patient with a lethal form metatropic dysplasia, dominant mutation (I604M) calcium channel gene TRPV4. When micromasses were grown chondrogenic differentiation conditions and compared control iPSCs, mutant TRPV4-iPSCs showed significantly...
Working with cultured dermal fibroblasts derived from control individuals and patients systemic sclerosis (SSc), we have examined the effects of protein kinase C-delta (PKC-delta) on type I collagen biosynthesis steady-state levels COL1A1 COL3A1 mRNAs. Rottlerin, a specific inhibitor PKC-delta, exerted powerful, dose-dependent inhibition III gene expression in normal SSc cells. Optimal rottlerin concentrations caused 70-90% production, >80% reduction mRNA, >70% mRNA both cell types. In vitro...
We recently reported the isolation and sequencing of two classes human alpha 2(VI) collagen cDNA clones which share common sequences for first two-thirds molecule but contain a different sequence either 607 or 887 base pairs at their 3' ends (Chu, M.-L., Pan, T.-C., Conway, D., Kuo, H.-J., Glanville, R. W., Timpl, R., Mann, K., Deutzmann, (1989) EMBO J. 8, 1939-1946). In present study, we report another clone, is identical to one class previously isolated cDNAs except 293-base pair insertion...
