Significance Epigenetic regulations control the accessibility of transcription factors to their target regions. Modulation chromatin determines which transcripts be expressed and therefore, defines cell identity. Chromatin modulation during fate determination involves a complex regulatory network, yet comprehensive view remains explored. Here, we provide global muscle stem activation. We identified long noncoding RNA (lncRNA), LncMyoD , regulates lineage progression through modulating...

T helper type 2 (Th2) cytokines, such as interleukin (IL)-4, IL-5, and IL-13, are the central therapeutic targets in development of drugs against asthma. Recently, it was found that bavachinin, an essential small molecule Psoralea corylifolia, has very efficient function asthma treatment a murine model via selective inhibition Th2 cytokine production. However, clinical potential bavachinin is limited by its extremely low water solubility (<30 ng/mL). Here, we fabricated nanoparticle delivery...

Functional depletion of the U1 small nuclear ribonucleoprotein (snRNP) with a 25 nt AMO (antisense morpholino oligonucleotide) may lead to intronic premature cleavage and polyadenylation thousands genes, phenomenon known as snRNP telescripting; however, underlying mechanism remains elusive. In this study, we demonstrated that could disrupt structure both in vitro vivo, thereby affecting snRNP–RNAP polymerase II interaction. By performing chromatin immunoprecipitation sequencing for...

It has recently been shown that CFIm25, a canonical mRNA 3' processing factor, could play variety of physiological roles through its molecular function in the regulation alternative polyadenylation (APA). Here, we used CRISPR/Cas9-mediated gene editing approach human embryonic stem cells (hESCs) for and obtained three knockdown/mutant cell lines. CFIm25 resulted higher proliferation rate impaired differentiation potential hESCs, with these effects likely to be directly regulated by target...

ABCA7 encodes an ATP-binding cassette transporter, and its loss-of-function variants are associated with Alzheimer's disease. To investigate the role of deficiency in pathogenesis disease, we generated a homozygous ABCA7-knockout induced pluripotent stem cell (iPSC) line using CRISPR/Cas9-mediated gene editing. This ABCA7-deficient iPSC maintains normal karyotype, expression pluripotency markers, trilineage differentiation capacity.

Alzheimer's disease (AD) is known as a leading cause of dementia in elderly persons. It chronic neurodegenerative disorder characterized by progressive cognitive dysfunction. AD can disrupt functional connectivity distributed cortical networks. The S-estimator, which measure multivariate intraregional synchronization, was analyzed this study. Twenty patients with and 20 age-matched controls were tested at baseline after 1 year to evaluate the potential synchronization be possible marker...

Abstract Functional depletion of U1 snRNP with a 25 nt AMO (antisense morpholino oligonucleotides) may lead to intronic premature cleavage and polyadenylation (PCPA) thousands genes, phenomenon known as telescripting; however, the underlying mechanism remains elusive. In this study, we demonstrated that could disrupt structure both in vitro vivo, thereby affecting snRNP/RNAP polymerase II (RNAPII) interaction. We further showed treatment might promote RNAPII disassociation pre-mRNA an RNA...
