Background This study assessed the long-term safety and efficacy of tiotropium Respimat, a long-acting inhaled anticholinergic bronchodilator, in asthma, added on to corticosteroids (ICS) with or without β2-agonist (LABA). Methods 285 patients symptomatic despite treatment ICS±LABA, were randomised 2:2:1 once-daily 5 μg, 2.5 μg placebo for 52 weeks (via Respimat SoftMist inhaler) double-blind, placebo-controlled, parallel-group (NCT01340209). Primary objective: describe profile tiotropium....
β-arrestins are critical signalling molecules that regulate many fundamental physiological functions including the maintenance of euglycemia and peripheral insulin sensitivity. Here we show inactivation β-arrestin-2 gene, barr2, in β-cells adult mice greatly impairs release glucose tolerance fed with a calorie-rich diet. Both KCl-induced secretion calcium responses were profoundly reduced (barr2) deficient β-cells. In human β-cells, barr2 knockdown abolished glucose-induced secretion. We...
Increased hepatic glucose production is a key pathophysiological feature of type 2 diabetes. Like all other cell types, hepatocytes express many G protein-coupled receptors (GPCRs) that are linked to different functional classes heterotrimeric proteins. The important physiological functions mediated by G(s)-coupled glucagon well-documented. In contrast, little known about the in vivo roles hepatocyte GPCRs proteins G(q) family. To address this issue, we established transgenic mouse line...
Abstract β-Arrestins are major regulators of G protein-coupled receptor-mediated signaling processes. Their potential roles in regulating adipocyte function vivo remain unexplored. Here we report the novel finding that mice lacking β-arrestin-2 (barr2) selectively adipocytes show significantly reduced adiposity and striking metabolic improvements when consuming excess calories. We demonstrate these beneficial effects due to enhanced through β3-adrenergic receptors (β3-ARs), indicating barr2...
Abstract Background & Aims Faldaprevir ( BI 201335) is a potent once‐daily QD ) NS 3/4A protease inhibitor for the treatment of patients with genotype‐1 GT ‐1) hepatitis C virus HCV ). The aim this study was to evaluate safety, pharmacokinetics and efficacy faldaprevir plus pegylated interferon alfa‐2a (Peg IFN ribavirin RBV in Japanese infected chronic ‐1 . Methods Part 1 phase II randomized, double‐blind, placebo‐controlled, dose‐ascending study. Treatment‐naïve received 120 or 240 mg...
A better understanding of the signaling pathways regulating adipocyte function is required for development new classes antidiabetic/obesity drugs. We here report that mice lacking β-arrestin-1 (barr1), a cytoplasmic and nuclear protein, selectively in adipocytes showed greatly impaired glucose tolerance insulin sensitivity when consuming an obesogenic diet. In contrast, transgenic overexpressing barr1 were protected against metabolic deficits caused by high-calorie Barr1 deficiency led to...
1. Testosterone cyclopentylpropionate shows the greatest anabolic and androgenic efficacy of agents studied. Its increased cannot be explained on basis delayed absorption, suggesting a inactivation this ester. 2. Aluminum phosphate delays onset decline actions testosterone in aqueous suspensions, but produces no such effect upon methylandrostenediol.
We have investigated the prevalence of dry mouth among patients with autoimmune diseases other than Sjögren's syndrome. One hundred and forty-four patients, excluding primary syndrome, were enrolled in this study. The volume saliva secreted was measured screening technique for estimation salivary flow, which uses a filter paper diagnosing mouth. Disturbed secretion observed 84 (58.3 %) 144 patients. In case free disturbed differed significantly disease groups (P < 0.05), being over 50 % all...
The efficacy and safety of nintedanib in patients with IPF were assessed the two replicate Phase III placebo-controlled INPULSIS trials. In both trials, reduced disease progression by reducing decline forced vital capacity (FVC). recommended dose was 150 mg twice daily (bid), but reductions to 100 bid treatment interruptions allowed for management adverse events. Following a reduction, could be re-escalated bid. We whether and/or influenced effect on FVC decline.
Abstract We performed laparoscopic Lich–Gregoir antireflux plasty on 4 patients with primary vesicoureteral reflux. All procedures were conducted using the extraperitoneal approach. The average surgical time was 230 min. There no complications. After surgery, voiding cysturethrograms showed reflux in all patients.
Liquid fibrin sealants, together with sheet-type hemostatic agents, have been used during partial nephrectomies to secure effective hemostasis at the suture site. Using animal kidneys, we investigated which agent might adhere most effectively renal tissue and serve best as a bolster. sealant alone, or in combination hemostat, such collagen, gelatin oxidized-cellulose was applied cut surface of kidney anesthetized rabbits, differences degree adherence resultant efficacy were evaluated....
