The Keap1-Nrf2 system plays a central role in cytoprotection against electrophilic/oxidative stresses. Although Cys151, Cys273, and Cys288 of Keap1 are major sensor cysteine residues for detecting these stresses, it has not been technically feasible to evaluate the functionality Cys273 or Cys288, since mutants that harbor substitutions maintain ability repress Nrf2 accumulation do exist. To overcome this problem, we systematically introduced amino acid into Cys273/Cys288 finally identified...

The Keap1-Nrf2 system plays a central role in the oxidative stress response; however, identity of reactive oxygen species sensor within Keap1 remains poorly understood. Here, we show that mutant lacking 11 cysteine residues retains ability to target Nrf2 for degradation, but it is unable respond cysteine-reactive inducers. Of mutated residues, find 4 (Cys226/613/622/624) are important sensing hydrogen peroxide. Our analyses multiple mice lines, complemented by MEFs expressing series mutants,...

We elucidated the metabolism of methylglyoxal (MG) in chloroplasts higher plants. Spinach showed MG-dependent NADPH oxidation because aldo-keto reductase (AKR) activity. K(m) for MG and V(max) AKR activity were 6.5 mm 3.3 µmol (mg Chl)(-1) h(-1) , respectively. Addition to illuminated induced photochemical quenching (Qp) Chl fluorescence, indicating that stimulated photosynthetic electron transport (PET). Furthermore, enhanced light-dependent uptake O(2) into chloroplasts. After illumination...

Numerous small molecules (termed inducers), many of which are electrophiles, upregulate cytoprotective responses and inhibit pro-inflammatory pathways by activating nuclear factor-erythroid 2 p45-related factor (NRF2). Key to NRF2 activation is the ability chemically modifying critical sensor cysteines in main negative regulator NRF2, Kelch-like ECH-associated protein 1 (KEAP1), C151, C273 C288 best characterized. This study aimed establish requirement for these cysteine sensor(s) biological...

β-Stereoselective mannosylation using donors bearing the 2,6-lactone moiety is described. In general, glycosylation a nucleophilic substitution reaction between an alcoholic nucleophile and sugar containing leaving group at anomeric position. Owing to stereoelectronic effects, tends proceed via SN1 mechanism afford α-glycosides. We found that introduction of bridge can circumvent competing reaction, affording β-glycosides with stereoinversion SN2(-like) mechanisms. Glycosyl...

In Arabidopsis thaliana, the aldo-keto reductase (AKR) family includes four enzymes (The AKR4C subfamily: AKR4C8, AKR4C9, AKR4C10, and AKR4C11). AKR4C8 AKR4C9 might detoxify sugar-derived reactive carbonyls (RCs). We analyzed AKR4C10 AKR4C11, compared enzymatic functions of enzymes. Modeling protein structures based on known structure found an (α/β)8-barrel motif in all Loop (A, B, C) which determine substrate specificity, differed among four.
