We have investigated the relationship in human cultured normal and malignant melanocytes between accumulation of mRNAs encoding tyrosinase tyrosinase‐related protein‐1 (TRP‐1), activity presence melanin. Tyrosinase mRNA correlates with pheomelanin, eumelanin or both melanin types. In contrast TRP‐1 is only detectable cells containing eumelanin, which suggests a role for synthesis pathway.
Galectin-3 (Gal-3) is a member of the family beta-galactoside-binding mammalian lectins, and has been implicated in tumour invasion metastatic process vitro vivo.To determine whether an increase serum Gal-3 production could be found patients with advanced melanoma. We collected 18 sera from AJCC stage IV melanomas 20 healthy volunteers. Determination was performed by ELISA, group melanoma, these results were compared lactate dehydrogenase (LDH) C-reactive protein (CRP) concentrations.Gal-3...
Clinical outcome of patients with high-risk melanoma cannot be reliably predicted on the basis classical histopathological examination. Our study aimed to determine in metastases a gene expression profile associated patient survival, and identify validate marker(s) poor clinical outcome. Skin lymph node from (training population) were used candidate prognostic based DNA microarray analysis. Additional skin (validation assess value first ranked by real-time PCR. We performed analysis training...
The purpose of this study was to compare the invasive properties normal human cutaneous melanocytes and a melanoma cell line (HBL) in three-dimensional model reconstructed skin. Specifically, we asked what extent pigmentary behaviour both cells is influenced by their interaction with adjacent skin (keratinocytes fibroblasts) basement membrane (BM). In presence BM, within remained basal layer keratinocytes did not pigment spontaneously. When BM removed, were found suprabasally pigmented...
The expression of E-selectin induced by tumor necrosis factor (TNF) on the surface human umbilical vein endothelial cells (HUVEC) was partially inhibited an increase in level adenosine 3',5'-cyclic monophosphate (cAMP), produced forskolin or cholera toxin combined with type IV phosphodiesterase inhibitor rolipram and protein kinase A agonist phosphorothioate analogue cAMP SpcAMPS. same agents had no significant effect constitutive TNF-stimulated intercellular adhesion molecule 1 (ICAM-1),...
α-Melanocyte stimulating hormone (α-MSH) is known to have pleiotrophic functions including pigmentary, anti-inflammatory, antipyretic and immunoregulatory roles in the mammalian body. It also reported influence melanoma invasion with levels of α-, β- γ-MSH correlated clinically malignant development, but other studies suggest α-MSH acts retard invasion. In present study, we investigated action on three human cell lines (HBL, A375-SM C8161) differing metastatic potential....
We previously demonstrated an inverse correlation between tyrosinase-related protein 1 (TYRP1) mRNA expression in melanoma metastases and patient survival. However, TYRP1 was not detected half of tissues expressing did correlate with Based on a study reporting that 3′ untranslated region (UTR) contains two miR-155-5p (named miR-155) binding sites exhibiting single-nucleotide polymorphisms (SNPs) promote (matched miRNA–mRNA interaction) decay or (mismatched), we aimed to investigate the role...
Abstract Wound healing is facilitated by neoangiogenesis, a complex process that essential to tissue repair in response injury. MicroRNAs are small, noncoding RNAs can regulate the wound including stimulation of impaired angiogenesis associated with type‐2 diabetes (T2D). Expression miR‐409‐3p was significantly increased nonhealing skin wounds patients T2D compared non‐wounded normal skin, and murine model T2D. In high glucose, neutralization markedly improved EC growth migration human...

Alpha-MSH, a proopiomelanocortin (POMC)-derived peptide, is known to be produced in the pituitary, skin, and melanoma tumors possess many biological effects, mainly on melanocyte pigmentation growth. Moreover, expresses adhesion molecules, including ICAM-1. The latter has been reported play role spread associated metastatic process. We conducted study order evaluate possible effect of MSH ICAM-1 expression human cultured malignant normal melanocytes. Our data show that alpha-MSH inhibits...
Clinical outcome of high-risk melanoma patients is not reliably predicted from histopathological analyses primary tumours and often adjusted during disease progression. Our study aimed at extending our previous findings in skin metastases to evaluate the prognostic value tyrosinase-related protein 1 (TYRP1) lymph node stages III IV patients. TYRP1 mRNA expression 104 was quantified by real-time PCR normalised S100 calcium-binding B (S100B) correct for tumour load. TYRP1/S100B ratios were...
Angiogenesis is critical for tissue repair following myocardial infarction (MI), which exacerbated under insulin resistance or diabetes. MicroRNAs are regulators of angiogenesis. We examined the metabolic regulation miR-409-3p in post-infarct was increased patients with acute coronary syndrome (ACS) and a mouse model MI. In endothelial cells (ECs), induced by palmitate, while vascular growth factor (VEGF) fibroblast (FGF) decreased its expression. Overexpression EC proliferation migration...
Epidermal-type fatty acid-binding protein (E-FABP), a related to the intracellular trafficking of acids, is expressed in melanocytic tumours but not normal human melanocytes. E-FABP interacts with S100A7. The presence these two proteins was investigated urine patients cutaneous melanoma or other types cancer, and healthy controls. first voided morning samples 31 melanoma, 73 cancer 17 controls were concentrated submitted sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE)...
Kinnaert, E., Morandini, R., Simon, S., Hill, H. Z., Ghanem, G. and Van Houtte, P. The Degree of Pigmentation Modulates the Radiosensitivity Human Melanoma Cells. The relationship between cell pigmentation radiosensitivity was investigated in two selected human melanoma lines with different melanin content (mixed type: eumelanin pheomelanin, pheomelanotic phenotypes). same study also done after stimulation melanogenesis (1) by addition precursor l-tyrosine to each separately (2) irradiation...
Glutathione (GSH) and its precursor cysteine (Cys) are both known to react within any cells with oxidative species thus play an important role in cellular defense mechanisms against stress. In melanocytes, these also precursors of melanogenesis by reacting non-enzymatically l-dopaquinone form the sulfur-containing pheomelanin. Our aim was assess pigment radioprotection mechanism using a human melanoma cell model mixed-type melanin under GSH depletion obtain radiosensitizing effect. The...
Abstract Four α‐MSH drug conjugates have been synthesized, 2 C‐terminal (Pep 3 and 4) central fragments I 2), the latter being 4–10 sequence known to be main α‐MSH‐receptor‐recognition site. Melphalan was introduced into each at different locations. Their ability recognize receptors as well their cytotoxic effects were compared in cell lines: melanoma, carcinoma fibroblast cells. Pep able specifically bind MSH on melanoma cells by displacing labelled a‐MSH from its binding sites...
Seven human melanoma metastases were extracted in order to check the possible presence of any α‐melanocyte stimulating hormone (MSH) immunoreactivity. The aim that study was provide some explanation for, mainly, two observations we have already made and reported: 1) increased plasma α‐MSH levels tumour‐bearing patients as compared with tumour‐free patients; 2) specific receptors on cells culture. We could measure large amounts immunoreactive all tumours ranging from 0.31 4.27 pmoles per g...
