The mitochondrial, or intrinsic, apoptosis pathway is regulated mainly by members of the B-cell lymphoma 2 (BCL-2) protein family. BCL-2-associated X regulator (BAX) plays a pivotal role in initiation mitochondria-mediated as one factors causing mitochondrial outer-membrane permeabilization (MOMP). Of current interest are endogenous BAX ligands that inhibit its MOMP activity. Mitochondrial-derived peptides (MDPs) recently identified class retrograde signaling molecules and reported to be...

Hydroxyzine in doses of 1.2 - 20 mgm./kgm. intravenously caused a transient fall arterial blood pressure and inhibited the pressor response to epinephrine cats under pentobarbital anesthesia. Hydroxyzine reduced incidence duration ventricular arrhythmias treated with harman methosulfate epinephrine. Hydroxyzine increased coronary flow perfused cat's heart hearts dogs anesthesia. The femoral vessels were also dilated by hydroxyzine. Epinephrine-induced automaticity papillary muscles...

Members of the B-cell lymphoma (BCL-2) protein family regulate mitochondrial outer membrane permeabilization (MOMP), a phenomenon in which mitochondria become porous and release death-propagating complexes during early stages apoptosis. Pro-apoptotic BCL-2 proteins oligomerize at MOMP, inducing pore formation. Of current interest are endogenous factors that can inhibit pro-apoptotic translocation oligomerization. A mitochondrial-derived peptide, Humanin (HN), was reported being expressed...

Presented here is a novel method for encapsulating proteins into biodegradable, thermoresponsive coacervate-type polyesters. Bovine serum albumin (BSA) was efficiently incorporated coacervate droplets via simple encapsulation mechanism. Tunable modular systems such as the one presented may be useful in range of protein delivery applications.

Mitochondrial-derived peptides are encoded by mitochondrial DNA but have biological activity outside mitochondria. Eight of these sequences within the 12S and 16S ribosomal genes: humanin, MOTS-c, six SHLP peptides, SHLP1-SHLP6. These various effects in cell culture animal models, affecting neuroprotection, insulin sensitivity, apoptosis, some secreted, potentially having extracellular signaling roles. However, except for their importance normal function is unknown. To gauge importance,...

α-Amidinoazadi(benzopyrro)methenes were synthesized using the Re(CO)3 unit as a templating agent. The products of these template reactions are six-coordinate rhenium complexes, with facial arrangement carbonyls, noncoordinating anion, and tridentate α-amidinoazadi(benzopyrro)methene ligand. ligand shows conversion one diiminoisoindoline sp2 carbon to sp3 carbon, which has been seen in "helmet" bicyclic phthalocyanines. bidentate fragment tilts out plane coordination. Selective hits for the glutaredoxin ortholog of Brucella melitensis are determined using STD NMR and verified by trNOE (15)N-HSQC titration. The most promising hit, RK207, was docked into target molecule a scoring function to compare simulated poses experimental data. After elucidating possible poses, hit further optimized lead compound extension with an electrophilic acrylamide warhead. We believe that focusing on selectivity in this early stage drug discovery will limit cross-reactivity...
