Background . Excessive autophagy is a major mechanism of myocardial ischemia reperfusion injury (I/RI) in diabetes with enhanced oxidative stress. Antioxidant N-acetylcysteine (NAC) reduces I/RI. It unknown if inhibition may represent whereby NAC confers cardioprotection diabetes. Methods and Results Diabetes was induced Sprague-Dawley rats streptozotocin they were treated without or (1.5 g/kg/day) for four weeks before being subjected to 30-minute coronary occlusion 2-hour reperfusion. The...

Background: Telomere shortening and dysfunction may cause metabolic disorders, tissue damage age-dependent pathologies. However, little is known about the association of telomere-associated protein Rap1 with mitochondrial energy metabolism cardiac aging. Methods: Echocardiography was performed to detect structure function in Rap1+/+ Rap1-/- mice at different ages (3 months, 12 months 20 months). length, DNA damage, senescence cardiomyocyte size were analyzed using real-time PCR, Western...

Abstract Aims/Introduction Diabetic hearts are more vulnerable to ischemia‐reperfusion injury (I/RI). The activation of nucleotide‐binding oligomerization domain‐like receptor protein 3 (NLRP3) inflammasome can mediate the inflammatory process, and hence might contribute myocardial I/RI. Activation autophagy eliminate excess reactive oxygen species alleviate I/RI in diabetes. present study aimed investigate whether diabetic through inhibition NLRP3 activation. Materials Methods A dose 65...

The mitochondrial, or intrinsic, apoptosis pathway is regulated mainly by members of the B-cell lymphoma 2 (BCL-2) protein family. BCL-2-associated X regulator (BAX) plays a pivotal role in initiation mitochondria-mediated as one factors causing mitochondrial outer-membrane permeabilization (MOMP). Of current interest are endogenous BAX ligands that inhibit its MOMP activity. Mitochondrial-derived peptides (MDPs) recently identified class retrograde signaling molecules and reported to be...

Inflammation plays an important role in the pathogenesis of coronary artery calcification (CAC). This study aims to explore potential association between inflammation indices derived from complete blood count (CBC) and CAC, including neutrophil lymphocyte ratio (NLR), (dNLR), neutrophil-monocyte (NMLR), systemic response index (SIRI), immune-inflammation (SII), aggregate (AISI), platelet (PLR), monocyte (MLR). We systematically collected data patients who underwent CAC scoring via cardiac CT...

Calcium/calmodulin-dependent protein kinase II δ (CaMKIIδ) has a pivotal role in cardiac signaling. Constitutive and deleterious CaMKII "autonomous" activation is induced by oxidative stress, the previously reported mechanism involves oxidation of methionine residues regulatory domain. Here, we demonstrate that covalent leads to disulfide bond with Cys273 domain causing autonomous activity. Autonomous was treating diamide or histamine chloramine, two thiol-oxidizing agents. Autonomy reversed...

Lung ultrasound-guided alveolar recruitment manoeuvres (RMs) may reduce the lung ultrasound score. However, whether use of this strategy can incidence postoperative pulmonary complications (PPCs) in adult obese population has not yet been tested. This is a single-centre, two-arm, prospective, randomised controlled trial. A total number 476 patients scheduled for bariatric surgeries will be enrolled. They randomly assigned to receive either RMs (intervention group) or conventional (control...

Sirt1 is an NAD+-dependent protein deacetylase that regulates many physiological functions, including stress resistance, adipogenesis, cell senescence and energy production. can be activated by deprivation, but the mechanism poorly understood. Here, we report negatively regulated ATP, which binds to C-terminal domain (CTD) of Sirt1. ATP suppresses activity impairing CTD's ability bind as well its function substrate recruitment site. not NAD+, causes a conformational shift less compact...

Abstract Glia derived secretory factors play diverse roles in supporting the development, physiology, and stress responses of central nervous system (CNS). Through transcriptomics imaging analyses, we have identified Obp44a as one most abundantly produced proteins from Drosophila CNS glia. Protein structure homology modeling Nuclear Magnetic Resonance (NMR) experiments reveal a fatty acid binding protein (FABP) with high affinity towards long-chain acids both native oxidized forms. Further...

While a significant component of atherosclerotic plaques has been characterized as amyloid, the specific proteins remain to be fully identified. Probable amyloidogenic are apolipoproteins (Apos), which vital for formation and function lipoproteins. ApoCIII is an abundant protein implicated in atherosclerosis, we show it forms ribbonlike looped strikingly similar that previously reported ApoAI ApoCII. Triangles squares with width ~50 nm were also observed, may novel form amyloid or related rings.
