Activator protein-1 (AP-1) is a transcription factor that consists of either Jun-Jun homodimer or Jun-Fos heterodimer. Transactivation AP-1 required for tumor promoter-induced transformation in mouse epidermal JB6 cells and progression human keratinocytes. Until now, the question whether transactivation carcinogenesis vivo has remained unanswered, as issue functionally significant target genes. To address these issues we have generated transgenic which mutant c-jun (TAM67), under control...

Natural Killer (NK)1.1+ (NK1) T cells are a specialized subset of alpha/beta that coexpress surface receptors normally associated with the NK cell lineage innate immune system. On recognition conserved, major histocompatibility complex class I-like CD1 molecule, these able to release explosive bursts interleukin 4 (IL-4), cytokine promotes helper type 2 (Th2) effector an response. A unique feature their receptor (TCR) repertoire is expression invariant TCR alpha chain, V 14-J 281, and...

Target cell expression of certain MHC class I molecules correlates with resistance to lysis by NK cells. To explain this correlation, one hypothesis states that cells may possess two types receptors; activate whereas another, specific for target molecules, inhibit natural killing transducing negative signals. The surface molecule, Ly-49, is expressed on an subpopulation (15% 20%) in spleens from C57BL/6 (H-2b) mice. Previously, we showed Ly-49+ IL-2-activated was globally inhibited when...

Transplacental passage of cells from mother to fetus during murine pregnancy was examined by using glucose phosphate isomerase (GPI) or fluorescein tagging as markers. Female mice strains (BALB/cCr X C3H/HeJ)/F1 (A/J C57BL/6J)F1 (both Gpi-1a/b) were mated the corresponding Gpi-la males A/J, respectively). Cells liver, blood, and/or spleen in offspring typed at days 15, 16, 17, 18 gestation, day delivery, 1 postpartum. Only two 172 Gpi-1a/a obtained these matings showed evidence maternal cell...

We generated transgenic mice expressing a single-chain beta2-microglobulin (beta2m)-H-2Dd. The cell-surface beta2m-H-2Dd molecule was expressed on beta2m-deficient background and reacted with appropriate mAbs. It of the expected m.w. directed normal development CD8+ T cells in thymus broad TCR repertoire. also presented both exogenously provided endogenous peptide Ags to effector cells. Abstract Mouse oocytes at the dictyate and metaphase II stages as well fertilized eggs have been studied by indirect immunofluorescence for expression of H‐2 histocompatibility antigens on surface membranes. Serologically specific reactivity to antibody was observed patchy fluorescence distributed over oocyte membrane. In contrast, one‐cell zygotes exhibited variable reactivity, early two‐cell were negative. Absorption studies confirmed serologic specificity oocytes, which could be shown due...

Resistance to thyroid hormone (RTH) is a genetic disease caused by mutations of the receptor beta gene (TRbeta). One symptoms in some affected individuals growth retardation. While Jun/Fos-containing transcription factors are known to be necessary for many TCR-mediated events in mature T cells, relatively little is about their roles thymocyte development. We have generated transgenic mice that express a trans-dominant-negative mutant of c-Jun (TAM-67) specifically thymocytes. Expression TAM-67 inhibited the up-regulation AP-1-responsive genes such as c-jun and IL-2 stimulated thymocytes from mice. In addition, altered development TAM-67-expressing was revealed by...

SUMMARY Mouse oocytes at the dictyate stage were tested for presence of H‐2 antigens and Ia antigens, by means indirect immunofluorescence. Oocytes reacted positively with NIH alloantisera, but negative anti‐Ia region alloantisera. Results monoclonal antibodies indicate very low antigen density on oocytes.
