Adult T-cell leukemia/lymphoma (ATLL) is an aggressive malignancy with a poor prognosis current therapy. Here we report genome-wide CRISPR-Cas9 screening of ATLL models, which identified CDK6, CCND2, BATF3, JUNB, STAT3, and IL10RB as genes that are essential for the proliferation and/or survival cells. As single agent, CDK6 inhibitor palbociclib induced cell cycle arrest apoptosis in models wild-type TP53. had inactivated TP53 genetically were relatively resistant to owing compensatory CDK2...

Adult T-cell leukemia/lymphoma (ATLL) is one of the aggressive peripheral neoplasms with a poor prognosis. Accumulating evidence demonstrates that escape from adaptive immunity hallmark ATLL pathogenesis. However, mechanisms by which cells evade natural killer (NK)-cell-mediated have been poorly understood. Here we show CD48 expression in determines sensitivity for NK-cell-mediated cytotoxicity against cells. We performed unbiased genome-wide clustered regularly interspaced short palindromic...

Data on the response to COVID-19 vaccine in patients with myeloid malignancy, who are at severe risk case of infection, have not emerged. In a study 69 malignancies, including 46 acute leukaemia (AML) and 23 myelodysplastic syndrome (MDS), anti-spike SARS-CoV-2 antibody titres were measured 3 months after second mRNA-based vaccination. Seroconversion rates for AML MDS 94.7% 100% respectively, no significant difference from healthy controls (HCs). Patients showed significantly lower titre...

Serologic responses of COVID-19 vaccine are impaired in patients with B-cell lymphoma, especially those who had recently been treated anti-CD20 monoclonal antibodies. However, it is still unclear whether develop an immune response following vaccination. We investigated the efficacy vaccination against SARS-CoV-2 171 non-Hodgkin lymphoma (B-NHL) received two doses mRNA-based and we compared to that 166 healthy controls. Antibody titers were measured 3 months after administration second dose....

Patients with hematologic diseases have experienced coronavirus disease 2019 (COVID-19) a prolonged, progressive course. Here, we present clinical, pathological, and virological analyses of three cases prolonged COVID-19 among patients undergoing treatment for B-cell lymphoma. These had all been treated anti-CD20 antibody bendamustine. Despite various antiviral treatments, high severe acute respiratory syndrome 2 (SARS-CoV-2) levels persisted >4 weeks, two them succumbed to COVID-19. The...

Abstract Intestinal microbiota plays an important role in the regulation of allogeneic immune reaction after hematopoietic stem cell transplantation (allo‐ SCT ). graft‐vs‐host disease ( GVHD ) is one major causes mortality allo‐ and often complicated with intestinal dysbiosis. Recent studies suggest that antibiotic‐induced dysbiosis a risk factor for . Summary Prolonged SARS‐CoV‐2 infection in immunocompromised individuals has been scattered, but the details remain unclear. We conducted a prospective study with 26 COVID‐19 patients haematological malignancies to determine viral shedding kinetics and characteristics. obtained nasopharyngeal swabs from 21–28 days post‐onset for PCR test performed virus isolation PCR‐positive samples. A viable was detected five (19.2%), all of whom had malignant lymphoma. Those significantly lower CD4 +...

It has been suggested that the effect of coronavirus disease 2019 (COVID-19) booster vaccination in patients with B-cell non-Hodgkin’s lymphoma (B-NHL) is inferior to healthy individuals. However, differences according histological subtype or treatment status are unclear. In addition, there less research on who subsequently develop breakthrough infections. We investigated effects first COVID-19 for B-NHL and clinical features infections Omicron variant era. this study, was classified into...

Benign precursors of B lymphocytes, termed hematogones, are observed in the regenerative state hematopoiesis following chemotherapy or allogeneic hematopoietic stem cell transplantation (allo-HSCT). Previous studies have demonstrated that expansion hematogones correlates with better clinical outcomes after allo-HSCT. We retrospectively analyzed association between and 309 consecutive patients who underwent allo-HSCT, which is largest population-based cohort reported so far. The incidence was...

We have reported that seroconversion rates after the second dose of mRNA-based COVID-19 vaccines for myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML) were 100% 95% respectively, with no significant difference from healthy controls (HCs).However, there are very limited data response to a third vaccine in those patients. In this complementary study, we investigated booster effect patients malignancies.A total 58 including 20 MDS 38 AML enrolled. Post‐mortem analysis revealed multiple tachyzoites T oxoplasma gondii in the ganglia,...

Human herpesvirus-6 (HHV-6) reactivation is sometimes observed in immunocompromised patients, especially after allogeneic stem cell transplantation. The complications of HHV-6 this setting are mainly recognized as encephalitis. We herein report the case a patient who developed pneumonitis cord blood transplantation (CBT). A 35-year-old male underwent CBT for T-cell/myeloid mixed phenotype acute leukemia and achieved neutrophil engraftment on day 31. He had received foscarnet prophylaxis...
