ABSTRACT Drug efflux is an important resistance mechanism in Mycobacterium tuberculosis . We found that verapamil, inhibitor, profoundly decreases the MIC of bedaquiline and clofazimine to M. by 8- 16-fold. This exquisite susceptibility was noted among drug-susceptible drug-resistant clinical isolates. Thus, inhibition sensitizer clofazimine, may emerge as a these drugs.
Mycobacterium tuberculosis is an important human pathogen, and yet diagnosis remains challenging. Little research has focused on the impact of M. gut microbiota, despite significant immunological homeostatic functions gastrointestinal tract. To determine effect infection we followed mice from aerosol until death, using 16S rRNA sequencing. We saw a rapid change in microbiota response to infection, with all showing loss then recovery microbial community diversity, found that pre-infection...
Next-Generation Sequencing (NGS) of 16S rRNA gene is now one the most widely used application to investigate microbiota at any given body site in research. Since NGS more sensitive than traditional culture methods (TCMs), many studies have argued for them replace TCMs. However, are we really ready this transition? Here compare diagnostic efficiency two using a large number samples (
Osmotolerance is one of the critical factors for successful survival and colonization microbes in saline environments. Nonetheless, information about these osmotolerance mechanisms still inadequate. Exploration soil microbiome its community structure novel genetic elements likely to provide on involved osmoadaptation. The present study explores native 16S rRNA gene sequence analysis has indicated dominance halophilic/halotolerant phylotypes affiliated Proteobacteria, Actinobacteria,...
Heterogeneity amidst healthy individuals at genomic level is being widely acknowledged. This, in turn, modulated by differential response to environmental cues and treatment regimens, necessitating the need for stratified/personalized therapy. We intend understand molecular determinants of Ayurvedic way (ancient Indian system medicine) endo-phenotyping into distinct constitution types termed "Prakriti", which forms basis personalized treatment. In this study, we explored analyzed human gut...
Antibiotic-resistant pathogens constitute an escalating public health concern. Hence a better understanding of the underlying processes responsible for this expansion is urgently needed. Co-selection heavy metal/biocide and antibiotic resistance genes (ARGs) has been suggested as one potential mechanism promoting proliferation antimicrobial (AMR). This paper aims to elucidate interplay exploit differences in usage infer patterns co-selection by non-antibiotic factors metals biocides context...
Studying respiratory illness-specific microbial signatures and their interaction with other micro-residents could provide a better understanding of lung ecology. Each illness has specific disease etiology, however, so far no study revealed disease-specific markers. The present was designed to determine features interactions residents in chronic obstructive pulmonary diseases (stable exacerbated), sarcoidosis, interstitial diseases. Broncho-alveolar lavage samples (n = 43) were analyzed by...
Growing up with siblings has been linked to numerous health outcomes and is also an important determinant for the developing microbiota. Nonetheless, research into role of having on microbiota mainly incidental.
Microbes are ubiquitously distributed in nature, and recent culture-independent studies have highlighted the significance of gut microbiota human health disease. Fecal DNA is primary source for majority microbiome studies. However, further improvement needed to obtain fecal metagenomic with sufficient amount good quality but low host genomic contamination. In current study, we demonstrate a quick, robust, unbiased, cost-effective method isolation high molecular weight (>23kb) (260/280 ratio...
Abstract Background From early life, children are exposed to a multitude of environmental exposures, which may be crucial importance for healthy development. Here, the microbiota particular interest as it represents interface between factors and child. As infants in modern societies spend considerable amount time indoors, we hypothesize that indoor bed dust might an important factor child colonization airway microbiome. To explore this hypothesis, analyzed influence exposures on 577 samples...
Bedaquiline is a newly approved drug for the treatment of multidrug-resistant tuberculosis, but there are concerns about its safety in humans. We found that coadministration verapamil with subinhibitory doses bedaquiline gave same bactericidal effect mice as did full human bioequivalent dosing. Adding to monotherapy also protected against development resistant mutants vivo. The adjunctive use may permit lower be used and thereby reduce dose-related toxicities tuberculosis patients.
Abstract Every niche in the biosphere is touched by seemingly endless capacity of microbes to transform world around them adapting swiftly and flexibly environmental changes, likewise gastrointestinal tract no exception. The ability cope with rapid changes external osmolarity an important aspect gut for their survival colonization. Identification these mechanisms a pivotal step towards understanding genomic suitability symbiont successful human Here we highlight our recent work applying...
Mycobacterium tuberculosis modulates levels and activity of key intracellular second messengers to evade protective immune responses. Calcium release from voltage gated calcium channels (VGCC) regulates responses pathogens. In this study, we investigated the roles VGCC in regulating immunity mycobacteria vitro vivo. Inhibiting L-type or R-type dendritic cells (DCs) either using antibodies by siRNA increased influx an inositol 1,4,5-phosphate activated channel dependent mechanism that...
Survival in a given environment requires specific functions, so genomic variation is anticipated within individual taxonomic groups that exhibit large diversity lifestyles. In this study, we sequence and assemble the genome of Oceanobacillus faecalis strain HM6, resident human gut. Using genus HM6 draft sequence, explore functional requirements for survival symbiotic arrangement gut, contrast to free living environment. Comparative genomics seven available complete genomes highlight...
The highly co-evolved relationship of parasites and their hosts appears to include modulation host immune signals, although the molecular mechanisms involved in host-parasite interplay remain poorly understood. Characterization these key genes cognate proteins related should lead a better understanding this intriguing biological phenomenon. malaria agent Plasmodium falciparum is predicted export cohort several hundred remodel erythrocyte. However, actively exported by asexual intracellular...
Antimicrobial peptides such as cathelicidins are important components of innate immune defence against inhaled microorganisms, and have shown antimicrobial activity Mycobacterium tuberculosis in vitro models. Despite this, little is known about the regulation expression cathelicidin during vivo. We sought to determine whether cathelicidin-related peptide gene (Cramp), murine functional homologue human (CAMP or LL-37), required for protective immunity M. infection used Cramp
A hallmark of protective immunity during Mycobacterium tuberculosis (M. tb) infection is the regulated secretion pro-inflammatory and regulatory cytokines. Suppressors Cytokine Signaling (SOCS) are key regulators cytokine function. In this study we investigated regulation Toll-like receptor 2 (TLR2) dendritic cell-specific ICAM-3 grabbing non-integrin 1 (DC-SIGNR1)-mediated SOCS1 expression in DCs M. tb infection. We show that, compared with TLR2, stimulating DC-SIGNR1 on induces higher...
Host-directed therapies that augment host immune effector mechanisms may serve as important adjunctive for tuberculosis treatment. We evaluated the activity of denileukin diftitox in an acute mouse model (TB) infection and analyzed cellular composition bacterial burden lungs spleens. These vivo studies show potentiates standard TB treatment model, effect which be due to depletion T-regulatory myeloid-derived suppressor cells during infection. Our results indicate other cell-depleting useful...
Major histocompatibility complex (MHC) molecules are a key element of the cellular immune response. Encoded by MHC they family highly polymorphic peptide receptors presenting antigens for surveillance T cells. We have shown that certain organic compounds can amplify responses catalyzing loading human class II HLA-DR. Here we show now achieve this interacting with defined binding site HLA-DR receptor. Screening compound library revealed set adamantane derivatives strongly accelerated rate....
