Toxoplasma gondii strains differ dramatically in virulence despite being genetically very similar. Genetic mapping revealed two closely adjacent quantitative trait loci on parasite chromosome VIIa that control the extreme of type I lineage. Positional cloning identified candidate gene ROP18 , a highly polymorphic serine-threonine kinase was secreted into host cell during invasion. Transfection virulent allele nonpathogenic III strain increased growth and enhanced mortality by 4 to 5 logs....

Host cell invasion in the Apicomplexa is unique its dependency on a parasite actin-driven machinery and exclusion of most host membrane proteins during parasitophorous vacuole (PV) formation. This occurs at junction between plasma membranes that has been called moving junction, circumferential zone which forms apical tip parasite, moves backward eventually pinches PV from membrane. Despite having described by electron microscopic studies 30 years ago, molecular nature this singular structure...

Toxoplasma gondii is an obligate intracellular parasite for which the discharge of apical organelles named rhoptries a key event in host cell invasion. Among rhoptry proteins, ROP2, prototype large protein family, translocated parasitophorous vacuole membrane during The ROP2 family members are related to protein-kinases, but only some them predicted be catalytically active, and none latter has been characterized so far. We show here that ROP18, member located re-localises at demonstrate...

Four rhoptry proteins (ROP) of Toxoplasma gondii previously identified with mAb have been affinity purified and analyzed by MS; the data obtained allowed genomic sequences to be assigned these proteins. As suggested for some them antibody crossreactivity, were shown belong a family, prototype which being ROP2. We describe here ROP2, 4, 5, 7. These four correspond most abundant products gene family that comprises several members we in EST libraries. Eight additional found cloned them. All...

Chronic HTLV-I (human T cell lymphotropic virus type I) infection may cause adult leukemia/lymphoma (ATL), a disease with dismal long-term prognosis. The transactivator, Tax, initiates ATL in transgenic mice. In this study, we demonstrate that an As2O3 and IFN-α combination, known to trigger Tax proteolysis, cures Tax-driven Unexpectedly, combination therapy abrogated initial leukemia engraftment into secondary recipients, whereas the primary tumor bulk still grew hosts, only ultimately...

Abstract Acute myeloid leukemia (AML) pathogenesis often involves a mutation in the NPM1 nucleolar chaperone, but bases for its transforming properties and overall association with favorable therapeutic responses remain incompletely understood. Here we demonstrate that an oncogenic mutant form of (NPM1c) impairs mitochondrial function. NPM1c also hampers formation promyelocytic (PML) nuclear bodies (NB), which are regulators fitness key senescence effectors. Actinomycin D (ActD), antibiotic...

Toxoplasma gondii, as many intracellular parasites, is separated from the cytosol of its host cell by a parasitophorous vacuole membrane (PVM). This forms during invasion and parasite apical organelles named rhoptries discharge proteins that associate with this process. It caused by Kaposi sarcoma-associated herpesvirus/human herpesvirus type 8 (KSHV/HHV8). occurs mainly, but not exclusively, in HIV-positive patients. PEL predominantly develops serous cavities and occasionally extracavitary regions. carries very poor prognosis with median survival time <6 months.
