Nanogels based on the polymers of galactosylated chitosan-graft-poly (N-isopropylacrylamide) (Gal-CS-g-PNIPAm) were used as carriers oridonin (ORI) for tumor targeting. Three ORI-loaded nanogels with various degrees galactose substitution prepared, and their characteristics evaluated. The release behavior ORI from these was pH-dependent, could be accelerated under mildly acidic conditions. cytotoxicity pH-sensitive. exhibited a higher antitumor activity than drug-loaded without...

Combining luminescent transition metal complex with super-resolution microscopy is an excellent strategy for the long-term visualization of dynamics subcellular structures in living cells. However, it remains unclear whether iridium(III) complexes are applicable a particular type technique, structured illumination (SIM), to image structures. Herein, dye, track mitochondrial cells under SIM described. The dye demonstrates specificity and photostability satisfactory cell permeability. While...

Abstract Heparin is an important anticoagulant drug, and microbial heparin biosynthesis a potential alternative to animal-derived production. However, effectively using synthesis enzymes faces challenges, especially with recombinant expression of active heparan sulfate N -deacetylase/ -sulfotransferase. Here, we introduce the monosaccharide -trifluoroacetylglucosamine into Escherichia coli K5 facilitate sulfation modification. The Protein Repair One-Stop Service-Focused Rational Iterative...

O-GalNAc glycans (or mucin O-glycans) play pivotal roles in diverse biological and pathological processes, including tumor growth progression. Structurally defined are essential for functional studies but synthetic challenges their inherent structural diversity complexity have limited access to these compounds. Herein, we report an efficient robust chemoenzymatic modular assembly (CEMA) strategy construct structurally glycans. The key this is the convergent of cores 1-4 6 from three chemical...

Triple-negative breast cancer (TNBC) is an extremely aggressive subtype associated with a poor prognosis. At present, the treatment for TNBC mainly relies on surgery and traditional chemotherapy. As key component in standard of TNBC, paclitaxel (PTX) effectively inhibits growth proliferation tumor cells. However, application PTX clinical limited due to its inherent hydrophobicity, weak penetrability, nonspecific accumulation, side effects. To counter these problems, we constructed novel...

Colla corii asini (E'jiao), donkey-hide gelatin prepared by stewing and concentrating from Equus asinus L. donkey hide, is a traditional Chinese medicine preparation widely used in clinical hematic antanemic therapy China. The aim of the present study was to investigate potential anti-aging effect explore related mechanisms D-galactose (gal) induced aging model mice. mice were artificially subcutaneously injection with D-gal at dose 100 mg/kg·d for 8 weeks. simultaneously treated them once...

Hyaluronidases are a family of enzymes that degrade hyaluronic acid (hyaluronan, HA) and widely used in many fields. A hyaluronidase producing bacteria strain was screened from the air. 16S ribosomal DNA (16S rDNA) analysis indicated belonged to genus Bacillus, named as Bacillus sp. A50. This is first report which yields unsaturated oligosaccharides product like other microbial hyaluronate lyases. Under optimized conditions, yield A50 could reach up 1.5×104 U/mL, suggesting good producer...

A novel chemoenzymatic approach for the synthesis of disialyl tetrasaccharide epitopes found as terminal oligosaccharides GD1α, GT1aα, and GQ1bα is described. It relies on chemical manipulation enzymatically generated trisaccharides conformationally constrained acceptors regioselective enzymatic α2–6-sialylation. This strategy provides a new route easy access to their derivatives.
