Goat antibodies to pig lung angiotensin-converting enzyme (kininase II) were conjugated microperoxidase. Rat tissue, previously incubated with non-immune goat serum, was the antibody-microperoxidase conjugate and then H2O2 3,3-diaminobenzidine. Electron microscopy revealed reaction product on plasma membrane caveolae of endothelial cells, especially those capillaries venules. These results support hypothesis that angiotensin I bradykinin are metabolized by enzymes luminal surface pulmonary cells.

Angiotensin-converting enzyme can be measured by the rate of release 3H-labelled hippurate from p-[3H]benzoylglycylglycylglycine. The product is separable substrate extraction acidified reaction mixtures with ethyl acetate. Assay results for human serum angiotensin-converting obtained within 1.5 h receipt samples.
