Claudin-7, a member of the claudin family, is highly expressed in distal nephrons kidneys and has been reported to be involved regulation paracellular Cl(-) permeability cell cultures. To investigate role claudin-7 vivo, we generated knockout mice (Cln7(-/-)) by gene-targeting deletion method. Here report that Cln7(-/-) were born viable, but died within 12 days after birth. showed severe salt wasting, chronic dehydration, growth retardation. We found urine Na(+), Cl(-), K(+) significantly...

Background. Pancreatic islet transplantation has shown great success in the treatment of diabetic patients. However, required immunosuppressive therapy exposes patients to serious side effects. Methods. We have designed a novel five-component/three-membrane capsule and encapsulation system protect transplanted cells from immune attack while allowing influx molecules nutrients necessary for cell function/survival efflux desired cellular product, specifically insulin, making recipients...

Animal models exhibiting high homology with humans at the genetic and pathophysiological levels will facilitate identification validation of gene targets underlying asthma. In present study, a nonhuman primate model allergic asthma was developed by sensitizing cynomolgus monkeys to dust mite antigen. Sensitization elevated allergen-specific serum IgE IgG levels, peripheral blood mononuclear cells isolated from sensitized animals released IL-4, IL-5, IL-10, but not IFN-gamma. Aerosolized...

Radiation therapy continues to be an important therapeutic strategy for providing definitive local/regional control of human cancer. However, oncogenes that harbor driver mutations and/or amplifications can compromise efficacy. Thus, there is a need novel approaches enhance the DNA damage produced by ionizing radiation.A forward chemical genetic approach coupled with cell-based phenotypic screening several tumor cell lines was used identify entity (NCE) functioned as radiation sensitizer....

ABSTRACT Background and objective: The mechanism by which iodopovidone achieves pleurodesis is unknown. This study investigated whether as effective doxycycline in producing systemic corticosteroids diminish its efficacy. Methods: Four groups of seven New Zealand rabbits were assigned to the following intrapleural treatment groups: 2 mL 2% iodopovidone, 4% plus 0.8 mg/kg triamcinolone intramuscularly weekly 10 mL/kg mL. Pleural fluid was collected 24, 48 72 h after injections analysed for...

Significance Skin and soft tissue infections (SSTIs) account for a majority of visits to hospitals clinics in the United States are typically caused by Gram-positive pathogens. Recently, it was discovered that bacteria use unique pathway synthesize critical cellular cofactor heme. The divergence heme biosynthesis pathways between humans provides opportunity development new antibiotics targeting this pathway. We report here identification small-molecule activator coproporphyrinogen oxidase...

Metabolic changes have been extensively documented in neurodegenerative brain disorders, including Parkinson's disease and Alzheimer's (AD). Mutations the C. elegans swip-10 gene result dopamine (DA) dependent motor dysfunction accompanied by DA neuron degeneration. Recently, putative human ortholog of (MBLAC1) was implicated as a risk factor AD, disorder that, like PD, has associated with mitochondrial dysfunction. Interestingly, AD MBLAC1 arises subjects cardiovascular morbidity,...

Organ toxicity induced by chemotherapeutic drugs is a serious obstacle in the effective treatment of patients suffering from cancer and autoimmune disease. A strong association exists between pulmonary toxicity, particularly fibrosis, drugs. Attempts have been made to identify compounds capable suppressing fibrosis. In addition its erythropoietic activity, erythropoietin (EPO) has shown effects on nonhemopoietic cells. Therefore, we postulated that EPO may exert beneficial lung tissue during...

The relationship between allergen-induced ventilatory drive and bronchoconstriction was investigated in dust mite-sensitive cynomolgus macaques periodically exposed to low doses of aerosolized antigen for up 5.5 yr. Initially, the animals responded mite allergen at a concentration 350 arbitrary units (AU)/ml with simultaneous increases lung resistance (RL) respiratory rate (RR). With time, RL RR became differentially sensitive provocation. At end study period, 15 AU/ml doubled without...

Abstract Background An adult male galago ( Otolemur garnettii ) presented for fight wounds following pairing breeding. Treatment was symptomatic with recovery. Following resolution, the animal re‐presented and died, despite additional treatment. Methods Necropsy, histopathology, bacterial cultures, 16S RNA sequencing. Results A large intrathoracic/intra‐abdominal abscess due to Trueperella pyogenes found at necropsy. Conclusions T. should be considered in abscesses/wounds of galagos.
