The potential risk of a nanoparticle as medical application has raised wide concerns, and this study aims to investigate silver (AgNP)-induced acute toxicities, genotoxicities, target organs the underlying mechanisms.Sprague-Dawley rats were randomly divided into 4 groups (n = each group), AgNP (containing Ag nanoparticles released Ag+, 5 mg/kg), Ag+ (released from same dose AgNP, 0.0003 5% sucrose solution (vechicle control) cyclophophamide (positive control, 40 mg/kg) administrated...
The discovery of G-protein coupled receptor (GPCR) accessory proteins has fundamentally redefined the pharmacological concept GPCR signaling, demonstrating a more complex molecular basis for specificity on plasma membrane and impressionable downstream intracellular cascades. not only contribute to proper folding trafficking receptors, but also exhibit selectable preferences. melanocortin (MRAP1 MRAP2) as well activity-modifying (RAMPs) are two well-known single transmembrane partners...
Oncolytic virotherapy is a new and safe therapeutic strategy based on the inherent cytotoxicity of oncolytic viruses their ability to replicate spread within tumors in selective manner. In previous study, type herpes simplex virus 2 (oHSV-2, named OH2) was constructed treat human cancers. That study demonstrated that OH2 genetically biologically stable. Its antitumor activity maintained, even after passaging for >20 generations. To advance into clinical trial, systematic preclinical safety...
Melanocortin-4 receptor (MC4R) functions as a crucial neuroendocrine G protein-coupled (GPCR) in the central nervous system of mammals, displaying agonist-independent constitutive activity that is mainly determined by its N-terminal domain. We previously reported zebrafish MC4R exhibited much higher basal cAMP level comparison to mammalian MC4Rs. However, functional evolution activities chordate MC4Rs remains be elucidated. Here we cloned and compared from nine vertebrate species showed...
Real-time polymerase chain reaction (PCR) has become an increasingly important technique for gene expression profiling because it can provide insights into complex biological and pathological processes be used to predict disease or treatment outcomes. Although normalized data are necessary accurate estimation of mRNA levels, several pieces evidence suggest that the so-called housekeeping genes is not stable. This study aimed validate reference normalization real-time PCR in...
The inositol polyphosphate-5-phosphatase E (Inpp5e) gene is located on chromosome 9q34.3. enzyme it encodes mainly hydrolyzes the 5-phosphate groups of phosphatidylinositol (3,4,5)-trisphosphate (PtdIns (3,4,5) P3) and (4,5)-bisphosphate (4,5)P2), which are closely related to ciliogenesis embryonic neurodevelopment, through mechanisms that largely unknown. Here we studied role Inpp5e in during neurodevelopment using inositol-deficiency neural tube defects (NTDs) mouse cell models. Confocal...
Intensive efforts have been made to develop models of hRSV infection or disease using various animals. However, the limitations such as semi-permissiveness and short duration impeded their applications in both pathogenesis therapeutics development. Here, we present a mouse model based on Rag2 gene knockout CRISPR/Cas9 technology. Rag2−/− mice sustained high viral loads upon intranasal inoculation with hRSV. The average peak titer rapidly reached 1 × 109.8 copies/g 1c106 TCID50 nasal cavity,...
As a widely applied nanomaterial, silver nanomaterials (AgNMs) have increased public concern about their potential adverse biological effects. However, there are few related researches on the long-term toxicity, especially reversibility of AgNMs in vivo. In current study, this issue was tackled by exploring liver damage after an intravenous injection nanorods with golden cores (Au@AgNRs) and its recovery relatively long term (8 w). After administration Au@AgNRs into rats, Ag found to be...
Background: CPGJ701 is a recombinant humanized anti-human epidermal growth factor receptor-2 (HER2) monoclonal antibody-derivative of the cytotoxic agent maytansine (DM1) conjugate for treatment HER2-positive metastatic breast cancer. Tissue cross-reactivity (TCR) studies in complete panel normal human, cynomolgus monkey and Sprague-Dawley were performed to provide evidence selecting animal species use preclinical toxicity predicting primary target organs clinical adverse drug reactions...
Vascular endothelial growth factor receptor 2 (VEGFR2)/KDR plays a critical role in tumor growth, diffusion, and invasion. The amino acid sequence homology of KDR between mouse human the VEGF ligand-binding domain was low, thus WT mice could not be used to evaluate Abs against KDR, lack suitable model hindered both basic research drug developments. Using CRISPR/Cas9 technique, we successfully inserted different fragments coding into chromosomal Kdr exon 4 locus obtain an hKDR humanized that...
The melanocortin system consists of five G protein–coupled receptors (MC1R-MC5R), the bidirectional endogenous ligands (MSH and Agouti families), accessory proteins (MRAP1 MRAP2). Accumulative studies vertebrate species find high expression level 1 receptor (MC1R) in dermal melanocyte elucidate essential roles skin fur pigmentation, morphological background adaptation, stress response. diploid amphibian Xenopus tropicalis ( xt ) has been utilized as a fantastic animal model for embryonic...
Glucagon plays an important role in glucose homeostasis and amino acid metabolism. It regulates plasma levels which turn modulate glucagon secretion from the pancreatic α-cell, thereby establishing a liver-α-cell axis described recently. We reported previously that knock-in mice bearing homozygous V369M substitution (equivalent to naturally occurring mutation V368M human receptor, GCGR) led hypoglycemia with improved tolerance. They also exhibited hyperglucagonemia, pancreas enlargement...
Lymphoma is the third most common cancer diagnosed in children, and T-cell lymphoma has worst prognosis based on clinical observations. To date, a model with uniform penetrance not yet been developed. In this study, we generated p53 deficient mouse by targeting embryonic stem cells derived from C57BL/6J strain. Homozygous mice exhibited higher rate of spontaneous tumorigenesis, high occurrence (93.3%) malignant lymphoma. Because tumor models phenotypic consistency are currently needed,...

Results The 80 had abdominal pain above symptoms...
Abstract Varicella zoster virus (VZV) causes chicken pox and shingles is prevalent worldwide. Acyclovir penciclovir (and its prodrugs) are first-line treatments for VZV infections, but they not highly potent against resistance may arise in immunocompromised people on long-term therapy. HPMPC (cidofovir) active VZV, cidofovir approved treating diseases, nephrotoxic, orally bioavailable. Here, we present the synthesis evaluation of USC-373, a phosphonate prodrug with activity other DNA...
